Effects of cold acclimation and dsRNA injections on Gs1l gene splicing in Drosophila montana

Abstract Alternative splicing, in which one gene produce multiple transcripts, may influence how adaptive genes respond to specific environments. A newly produced transcriptome of Drosophila montana shows the Gs1-like (Gs1l) gene to express multiple splice variants and to be down regulated in cold acclimated flies with increased cold tolerance. Gs1l’s effect on cold tolerance was further tested by injecting cold acclimated and non-acclimated flies from two distantly located northern and southern fly populations with double stranded RNA (dsRNA) targeting Gs1l. Nivelrikko on etenkin ikääntyneeseen väestöön kohdistuva kipuja ja toiminnallisia rajoituksia aiheuttava nivelsairaus. Tutkimuksen tarkoituksena oli selvittää liikunnan vaikutuksia polvinivelruston laatuun ja seurata mahdollisia rustomuutoksia 12 kuukauden kuluttua harjoittelun lopettamisesta lievää polvinivelrikkoa sairastavilla 50-65 –vuotiailla naisilla. Lisäksi tarkoituksena oli tutkia harjoittelun vaikutuksia fyysiseen toimintakykyyn ja polven oireisiin sekä arvioida vaikutusten pysyvyyttä intervention jälkeen. Histone acetylation plays a key role in the regulation of gene expression. The chromatin structure and accessibility of genes to transcription factors is regulated by enzymes that acetylate and deacetylate histones. The Sin3A corepressor complex recruits histone deacetylases and in many cases represses transcription. Here, we report that SAP30L, a close homolog of Sin3-associated protein 30 (SAP30), interacts with several components of the Sin3A corepressor complex. We show that it binds to the PAH3/HID (Paired Amphipathic Helix 3/Histone deacetylase Interacting Domain) region of mouse Sin3A with residues 120–140 in the C-terminal part of the protein. Estradiol deficiency and skeletal muscle apoptosis: Possible contribution of microRNAs.

Background Menopause leads to estradiol (E2) deficiency that is associated with decreases in muscle mass and strength. Here we studied the effect of E2 deficiency on miR-signaling that targets apoptotic pathways. Methods C57BL6 mice were divided into control (normal estrous cycle, n = 8), OVX (E2 deficiency, n = 7) and OVX + E2 groups (E2-pellet, n = 4). Six weeks following the OVX surgery, mice were sacrificed and RNA isolated from gastrocnemius muscles. miR-profiles were studied with Next-generation sequencing (NGS) and candidate miRs verified using qPCR. Aging of the musculoskeletal system: How the loss of estrogen impacts muscle strength.

Skeletal muscle weakness occurs with aging and in females this is compounded by the loss of estrogen with ovarian failure. Estrogen deficiency mediates decrements in muscle strength from both inadequate preservation of skeletal muscle mass and decrements in the quality of the remaining skeletal muscle. Processes and components of skeletal muscle that are affected by estrogens are beginning to be identified.
