Measurement methodology for greenhouse gas emissions from storage of forest chips–A review

Abstract Research on greenhouse gas emission related to solid biofuels has focused mainly on the emissions from end use and the production chain. GHG emissions from the storage of forest chips have not received much attention in recent literature. In order for EU emission reduction targets to be fully understood, emissions from solid biofuel storage needs to be better described. Usually emissions from chip piles have been modelled using studies from organic waste composting but these two materials can differ appreciably; for example the C/N-ratio and moisture content. Preparation of fungal conidia impacts their susceptibility to inactivation by ethanol vapours.

Abstract A common protocol employed for the preparation of conidia employs flooding a fungal colony grown on semi-solid media under optimum conditions with an aqueous solution. In contrast, conidia produced in a natural environment are usually not hydrated when disseminated in air and can be produced under water stress. In order to simulate the latter conditions, cultures were grown at different water activities and conidia were dry-harvested on the lid by turning the dishes upside-down then gently tapping the bottom of the box. Digital Holographic Microscopy: A New Imaging Technique to Quantitatively Explore Cell Dynamics with Nanometer Sensitivity

In the first part of this chapter, we describe how the new concept of digital optics applied to the field of holographic microscopy has made it possible to quantitatively and accurately measure the phase retardation induced on the transmitted wavefront by the observed transparent specimen, allowing thus to develop a reliable and flexible digital holographic quantitative phase microscopy (DH-QPM). In the second part the most relevant DH-QPM applications in the field of cell biology are presented. Real-time acquisition of complex optical fields by binary amplitude modulation

We describe, through simulations and experiments, a real-time wavefront acquisition technique using random binary amplitude masks and an iterative phase retrieval algorithm based on the Fresnel propagator. By using a digital micromirror device, it is possible to recover an unknown complex object by illuminating with this set of masks and simultaneously recording the resulting intensity patterns with a high-speed camera, making this technique suitable for dynamic applications.

Physical compensation of phase curvature in digital holographic microscopy by use of programmable liquid lens.

Quantitative phase measurements obtained with digital holographic microscopes are strongly dependent on the optical arrangement of the imaging system. The nontelecentric operation provides phase measurements affected by a parabolic phase factor and requires numerical postprocessing, which does not always remove all the perturbation. Accurate phase measurements are achieved by using the imaging system in telecentric mode. Unfortunately, this condition is not accomplished when a commercial microscope is used as the imaging system. Phase-shifting Gabor holography.

We present a modified Gabor-like setup able to recover the complex amplitude distribution of the object wavefront from a set of inline recorded holograms. The proposed configuration is characterized by the insertion of a condenser lens and a spatial light modulator (SLM) into the classical Gabor configuration. The phase shift is introduced by the SLM that modulates the central spot (dc term) in an intermediate plane, without an additional reference beam. Experimental results validate the proposed method and produce superior results to the Gabor method.

Off-axis digital holographic multiplexing for rapid wavefront acquisition and processing

Off-axis holographic multiplexing involves capturing several complex wavefronts, each encoded into off-axis holograms with different interference fringe orientations, simultaneously, with a single camera acquisition. Thus, the multiplexed off-axis hologram can capture several wavefronts at once, where each one encodes different information from the sample, using the same number of pixels typically required for acquiring a single conventional off-axis hologram encoding only one sample wavefront. Superresolved common-path phase-shifting digital inline holographic microscopy using a spatial light modulator.

Common-path phase-shifting lensless holographic microscopy has been recently proposed as a novel approach capable of high numerical aperture imaging in a lensless digital inline holographic microscopy layout [Opt. Lett.35, 3919 (2010)]. Here we present proof-of-concept validation for improving the resolution limit imposed by diffraction in such a setup. This is accomplished by shifting the phase lens displayed at the spatial light modulator, which moves the illumination point source to different off-axis positions. Quantitative Phase Imaging in Microscopy Using a Spatial Light Modulator

In this chapter, we present a new method capable of recovery of the quantitative phase information of microscopic samples. Essentially, a spatial light modulator (SLM) and digital image processing are the basics to extract the sample’s phase distribution. The SLM produces a set of misfocused images of the input sample at the CCD plane by displaying a set of lenses with different power at the SLM device. The recorded images are then numerically processed to retrieve phase information.
