Predictive assessment of ochratoxin A accumulation in grape juice based-medium by Aspergillus carbonarius using neural networks
2009
Aims: To study the ability of multi-layer perceptron artificial neural networks (MLP-ANN) and radial-basis function networks (RBFNs) to predict ochratoxin A (OTA) concentration over time in grape-based cultures of Aspergillus carbonarius under different conditions of temperature, water activity (a(w)) and sub-inhibitory doses of the fungicide carbendazim. Methods and Results: A strain of A. carbonarius was cultured in a red grape juice-based medium. The input variables to the network were temperature (20-28 degrees C), a(w) (0 center dot 94-0 center dot 98), carbendazim level (0-450 ng ml(-1)) and time (3-15 days after the lag phase). The output of the ANNs was OTA level determined by liqui…
Characterization and disruption of the cipC gene in the ochratoxigenic fungus Aspergillus carbonarius
2013
Aspergillus carbonarius is considered the most important ochratoxin A (OTA) producing fungi among those causing OTA contamination in grapes and grape-derived products. CipC is a small protein with unknown function that was previously found to be highly up-regulated in an OTA producer strain of A. carbonarius in comparison to a non OTA producer strain. In this study, cipC was characterized and disrupted via Agrobacterium tumefaciens-mediated transformation in an ochratoxigenic A. carbonarius strain in order to study whether this gene has a role in OTA production. Sequence analysis indicated that the promoter region of cipC contains putative binding sites for transcription factors that regula…
Green Extraction of Polyphenols from Waste Bentonite to Produce Functional Antioxidant Excipients for Cosmetic and Pharmaceutical Purposes: A Waste-t…
2022
In an ever-growing perspective of circular economy, the development of conscious, sustainable and environmental-friendly strategies to recycle the waste products is the key point. The scope of this work was to validate the waste bentonite from the grape processing industries as a precious matrix to extract polyphenols by applying a waste-to-market approach aimed at producing novel functional excipients. The waste bentonite was recovered after the fining process and opportunely pre-treated. Subsequently, both the freeze dried and the so-called “wet” bentonites were subjected to maceration. PEG200 and Propylene Glycol were selected as solvents due to their ability to dissolve poly…
Phylogenetic relationships among Flavescence doree strains and related phytoplasmas determined by heteroduplex mobility assay and sequence of ribosom…
2003
Heteroduplex mobility assay (HMA) and DNA sequencing were performed on Flavescence doree (FD) phytoplasma strains and related phytoplasmas belonging to the elm yellows group. Part of the ribosomal RNA gene operon and a nonribosomal DNA region were utilized for phylogenetic analyses. Two FD strains, FD92 and FD-D, detected in France and Italy, respectively, were identical in both DNA fragments, confirming previous results. Other FD strains were all very similar and most closely resembled ALY, an Italian alder phytoplasma. Phytoplasmas associated with German Palatinate grapevine yellows were shown to form a distinct subcluster, also different from the elm yellows phytoplasma subcluster. Strai…
The New Life Of Exhausted Bentonite From The Grape Processing Industry: From Waste To Precious Resource To Produce Novel Raw Materials Enriched In Po…
Introduction: In an ever-growing perspective of circular economy, the development of conscious, sustainable and environmental-friendly strategies to recycle the waste products is the key point1. The exhausted bentonite, from the grape processing industries, could become a precious matrix to produce new pharmaceutical/cosmetic excipients enriched in functional polyphenols2. Materials & Methods: The exhausted bentonite recovered after the fining process (supplied by Bono&Ditta s.p.a., Campobello di Mazara TP, Italy) was stored at -20°C. Solid-liquid extractions on both freeze-dried and as-collected (wet) bentonite were carried out by using PEG200 or propylene glycol as solvents. The l…
Proteomic profiling of German Dornfelder grape berries using data-independent acquisition
2017
Grapevine is one of the most important fruit plants throughout the world. Sequencing of the grape genome in 2007 enabled in-depth analyses of the grape proteome. Whereas many studies addressed changes in proteomic composition of grapes during ripening, we focused on the proteome of mature grape berries from Dornfelder, a characteristic red wine grape for Germany. Current data-independent acquisition proteomics technology enables the analysis of proteomic compositions in a degree of accuracy that was unreachable only a few years ago. Using a label-free proteomics approach, we quantified 712 proteins in mature Dornfelder grape berries, of which 650 could be annotated by the Blast2GO software.…
Developement of new techniques of Optimal Control in Quantum Dynamics : from nuclear magnetic resonance to molecular physics
2011
The goal of this thesis is to apply the optimal control theory to the dynamics of quantum systems. The first part aim at introducing the tools of optimal control in quantum control which were initially developedin mathematics. This approch has been applied on different kinds of quantum system with small and largedimensions. The first part of this manuscript introduces the optimal control tools which are used with a pointof view suited to a public of physicists. In the second part these techniques are used to control the dynamics ofspins in NMR and MRI. The third part deals with the development of new iterative algorithms applied to thecontrol by laser fields of the rotational dynamics of lin…
Moscato Cerletti, a rediscovered aromatic cultivar with oenological potential in warm and dry areas
2021
Baron Antonio Mendola was devoted to the study of grapevine, applying ampelography and dabbling in crosses between cultivars in order to select new ones, of which Moscato Cerletti, obtained in 1869, was the most interesting. Grillo, one of the most important white cultivars in Sicily, was ascertained to be an offspring of Catarratto Comune and Zibibbo, the same parents which Mendola claimed he used to obtain Moscato Cerletti. Thus the hypothesis of synonymy between Moscato Cerletti and Grillo or the same parentage for both sets of parents needs to be verified. In the present study, historical documents were consulted and genetic analyses and ampelographic, agronomic and qualitative character…
Use of selected indigenous yeasts from racemes of Grillo grape variety to improve the production of sparkling base wine
2019
The production of sparkling wines in Sicily is increasing considerably. The most important oenological characteristics of high quality sparkling wines are high content of acidity and low pH. In Sicily, white grape varieties such as those of the cultivar Grillo are characterized by very low concentrations of malic and tartaric acids due to hot climate and reduced rainfall typical of the production area. On the other hand, a very large amount of raceme grapes, characterized by low pH and high content of tartaric and malic acids, is largely produced by Grillo varieties. Hence, the winemaking of racemes might represent a technological solution to increase the quality of the acids in the final w…
Plant Response to Saline-Water Irrigation in a Sicilian Vineyard
2012
This chapter presents results of a 3-year field investigation in a vineyard located in Sicily (Mazara del Vallo, Trapani) within the framework of the Project “Evolution of cropping systems as affected by climate change” (CLIMESCO). Soil-plant responses to two saline irrigation waters were determined by measuring soil hydrological characteristics, soil salinity, crop transpiration and stomatal conductance in field plots of a Sicilian vineyard. The results proved that crop transpiration (T r) and stomatal conductance (G s) were significantly affected by soil salinity conditions, expressed by electrical conductivity of soil saturation extract (ECe). Significant reductions in T r and G s were f…
