The first component of complement (C1) is a macromolecule consisting of three distinct subcomponents, C1q, C1r, and C1s. In regard to its production site and its role in phagocytic processes it was of interest to find out whether these different subcomponents could be detected in human biopsy material only as a complex in individual cells or whether C1 subcomponents could be found on different cells. To study this question, monospecific fluorescein-labelled anti-human-C1q IgG and monospecific rhodamine-labelled anti-human C1q IgG were used. Morphologic Damage and Changes of Intracellular Calcium-Binding Sites after Acute Noise Trauma in the Organ of Corti of the Guinea Pig

In guinea pigs, an acute acoustic trauma was created by 6 consecutive gunshots. The sound pressure at the ear drum was 156 +/- 4 dB, the frequency maximum was between 4 and 6 kHz. Sixty hours after the noise trauma, the animals were decapitated, and the cochleae were prepared for microscopic analysis of the resulting trauma to the organ of Corti. During the process of fixation, the potassium-pyroantimonate precipitation reaction was performed to localize calcium-binding sites. The pattern of cell morphology and the distribution of calcium-binding sites was compared to that of normal control animals. Immunoelectron microscopic localization of nitric oxide synthase III in the guinea pig organ of Corti

Nitric oxide synthase III (NOS III) was identified in the guinea pig cochlea on an ultrastructural level using a post-embedding immunolabeling procedure. Ultrathin sections of London Resin (LR) White-embedded specimens were incubated with various concentrations of a commercially available antibody to NOS III and the immunoreactivity visualized by a gold-labeled secondary antibody. Analysis of ultrathin sections of the organ of Corti in the second turn of the cochlea showed that NOS III could be localized in the endothelial cells of the blood vessels under the basilar membrane, which was comparable to its location in similar cells types in various biological systems. The mode of lymphocyte transendothelial migration in the postcapillary high endothelial venules (HEVs) of Peyer's patches during normal homing and acute inflammation in the guinea pig was studied. Mit intracellularen Mikroelektroden wurden von Vorhof- und Kammermuskulatur von Meerscheinchenherzen Membran- und Aktionspotentiale abgeleitet. Die akute Wirkung von Trijodthyronin (T3 10−6 und 10−5 g/ml) auf diese elektrophysiologischen Mesgrosen wurde untersucht; auserdem wurden Veranderungen der intracellularen K- und Na-Konzentrationen in vitro und in vivo gemessen.

Es wird uber die Untersuchung einer akuten Wirkung von T3 in vitro auf die Refraktarperiode und die Leitungsgeschwindigkeit des Meerschweinchenvorhofs berichtet. T3 verkurzte die Refraktarzeit geringfugig (5,2%). Bei vermindertem K-Gehalt der Badlosung oder nach Vorgabe von ACh oder DNP trat die Verkurzung der Refraktarzeit unter T3 wesentlich deutlicher in Erscheinung, und es konnte Flimmern ausgelost werden, obwohl die Substanzen vor der Gabe von T3 vollstandig ausgespult waren. Adrenalin hatte keinen Einflus auf den T3-Effekt. Die Leitungsgeschwindigkeit wurde durch T3 nicht verandert.
