An experimental comparative study of dexamethasone, melatonin and tacrolimus in noise-induced hearing loss.

The calcineurin inhibitor tacrolimus (TCR) and the pineal gland hormone and antioxidant melatonin (MLT) have been shown to possess otoprotective properties against noise-induced hearing loss (NIHL). In contrast, dexamethasone (DXM) was not effective as an otoprotective agent against NIHL. Further studies are needed to understand the exact molecular mechanisms involved.Exposure to noise pollution and use of audio devices for long periods of time at high volume is known to cause hearing loss or NIHL. A theoretical basis for the high-frequency performance of the outer hair cell’s receptor potential

The frequency response of the outer hair cell (OHC) was studied theoretically. An electrical model of the OHC was analyzed mathematically, taking into account the effect of its inherent voltage-dependent capacitance. It was found that the variations of the capacitance dependent on the membrane potential could enhance the high-frequency response of the OHC, so that its cutoff frequency could be extended into the audio range. It was found further that the enhancement of the frequency response of the OHC was strongly dependent on its resting potential and on the ratio of the maximum voltage-dependent capacitance to the membrane linear capacitance.

Immunoelectron-microscopic localization of synaptophysin in the organ of Corti of the guinea pig.

Synaptophysin has been localized previously in the mammalian cochlea at the light-microscopic level and in few reports by electron microscopy using either a preincubation procedure or the avidin-biotin reaction. Here we present results of the electron-microscopic analysis for postembedding immunoreactivity of synaptophysin in the cochlea of the guinea pig of LR-White-embedded samples. Strong synaptophysin immunoreactivity is located in the cytoplasm of the efferent nerve endings at the base of inner and outer hair cells. Endothelial nitric oxide synthase upregulation in the guinea pig organ of Corti after acute noise trauma.

Endothelial nitric oxide synthase (eNOS) upregulation was identified 60 h after acute noise trauma in morphologically intact cells of the reticular lamina in the organ of Corti of the guinea pig in the second turn of the cochlea. Using gold-coupled anti-eNOS antibodies and electron microscopy, it was shown that eNOS expression was upregulated in all cell areas and cell types except inner hair cells. Furthermore, eNOS was found in the organelle-free cytoplasm and in mitochondria of various cell types. Immunoelectron microscopic localization of nitric oxide synthase III in the guinea pig organ of Corti

Nitric oxide synthase III (NOS III) was identified in the guinea pig cochlea on an ultrastructural level using a post-embedding immunolabeling procedure. Ultrathin sections of London Resin (LR) White-embedded specimens were incubated with various concentrations of a commercially available antibody to NOS III and the immunoreactivity visualized by a gold-labeled secondary antibody. Efficacy of three drugs for protecting against gentamicin-induced hair cell and hearing losses

BACKGROUND AND PURPOSE Exposure to an ototoxic level of an aminoglycoside can result in hearing loss. In this we study investigated the otoprotective efficacy of dexamethasone (DXM), melatonin (MLT) and tacrolimus (TCR) in gentamicin (GM)-treated animals and cultures. EXPERIMENTAL APPROACH Wistar rats were divided into controls (treated with saline); exposed to GM only (GM); and three GM-exposed groups treated with either DXM, MLT or TCR. Postnatal Neurogenesis and Neuronal Regeneration

The discovery of neurogenesis in the adult brain has challenged one of the central dogmas of neuroscience. Pioneer reports in rodents seed the ground for a detailed description in birds and reptiles, which was finally confirmed in discrete regions of several mammalian species including humans. This neurogenetical capability may serve as the basis for neuronal regeneration, as has already been described in the reptilian brain, and thus may represent a promising therapeutic approach. Consequently, in the last years there has been an important effort to deepen our knowledge of the biology, the functional significance and the regulation of adult neurogenesis.
