AbstractTherapeutic strategies combining the induction of effective antitumor immunity with the inhibition of the mechanisms of tumor-induced immunosuppression represent a key objective in cancer immunotherapy. Herein we demonstrate that effector/memory CD4+ T helper-1 (Th-1) lymphocytes, in addition to polarizing type-1 antitumor immune responses, impair tumor-induced CD4+CD25+FoxP3+ regulatory T lymphocyte (Treg) immunosuppressive function in vitro and in vivo. Th-1 cells also inhibit the generation of FoxP3+ Tregs from naive CD4+CD25−FoxP3− T cells by an interferon-γ–dependent mechanism. Abstract IL-9 has lent its numerical designation to the Th9 subset of CD4+ Th cells, although it is also produced by additional cell types, including mast cells. It is a pleiotropic cytokine involved in allergic reactions, parasitic infections, autoimmune inflammation, and cancer immunity. In this article, we provide evidence that NFATc2 has contradictory functions in the expression of IL-9 in murine Th9 cells and bone marrow–derived mast cells (BMMC). The basis for this is our observation that the production of IL-9 in NFATc2-deficient Th9 cells is increased, whereas it is decreased in BMMC devoid of NFATc2. Therapeutic Alliance: What Can be Learned from Case Studies?

Two different video-taped short-term psychotherapies were rated on therapeutic alliance for 27 sessions. The ratings were based on two methods: the Penn Helping Alliance Scales (Penn-HAS) and the Therapeutic Alliance Rating Scale (TARS). Both scales provide separate scores for the therapist and the patient. After checking the reliabilities of the ratings, the strength and course of the alliance formation are compared within and across therapies. Furthermore it was hypothesized that the alliance is structured differently in the two therapies.
