Electrochemical probe for the monitoring of DNA-protein interactions.

Self-assembly of thiol-terminated oligonucleotides on gold substrates provides a convenient way for DNA-functionalized surfaces. Here we describe the development of an electrochemical assay for the detection of DNA-protein interactions based on the modification of the electrochemical response of methylene blue (MB) intercalated in the DNA strands. Using a functionalized electrode with double stranded DNA carrying T3 RNA polymerase binding sequence, we show a substantial attenuation of the current upon the DNA-protein interaction. Moreover, a Langmuir binding isotherm for T3 RNA polymerase (T3 Pol) gives a dissociation constant K(D) equal to 0.46+/-0.23 microM. Such value is 100 times lower …

Label-free piezoelectric biosensor for prognosis and diagnosis of Systemic Lupus Erythematosus

[EN] An autoantigen piezoelectric sensor to quantify specific circulating autoantibodies in human serum is developed. The sensor consisted on a quartz crystal microbalance with dissipation monitoring (QCM-D) where TRIM21 and TROVE2 autoantigens were covalently immobilized, allowing the selective determination of autoantibodies for diagnosis and prognosis of Systemic Lupus Erythematosus (SLE). The sensitivity of the biosensor, measured as IC50 value, was 1.51 U/mL and 0.32 U/mL, for anti-TRIM21 and anti-TROVE2 circulating autoantibodies, respectively. [EN] Based on the unique ability of defibrillated sepiolite (SEP) to form stable and homogeneous colloidal dispersions of diverse types of nanoparticles in aqueous media under ultrasonication, multicomponent conductive nanoarchitectured materials integrating halloysite nanotubes (HNTs), graphene nanoplatelets (GNPs) and chitosan (CHI) have been developed. The aim of this study was to evaluate the effect of the inoculum to substrate ratio (ISR) and the mixture ratio between organic fraction of municipal solid waste (OFMSW) and sewage sludge (SS) on the methane production potential achievable from anaerobic co-digestion (AcoD). Biochemical Methane Potential (BMP) assays at mesophilic temperature were used to determine the best AcoD configuration for maximizing methane yield and production rate, as well as to address possible synergistic effects. Abstract Background: For the laboratory diagnosis of the antiphospholipid syndrome (APS) we developed a biosensor with the ability to distinguish between disease-relevant anti-β2-glycoprotein I (β2GPI) autoantibodies (anti-β2GPI) and pathogen-specific β2GPI cross-reactive antibodies that occur transiently during infections. Sono state valutate le performance di processo in termini di rimozione di TPH (86% con velocità di rimozione massima di 36 mgTPH kg-1 d-1) congiuntamente ad altre tipologie di indagini volte ad ottenere maggiori informazioni sulla natura dei processi che si sono sviluppati nel reattore. Leaching of sludge-borne trace elements has been observed in experimental and field studies. The role of microbial processes in the mobilization of trace elements from wastewater sludge is poorly defined.
