Multipolar interactions in the D pocket of thrombin: large differences between tricyclic imide and lactam inhibitors.

Two series of tricyclic inhibitors of the serine protease thrombin, imides (+/-)-1-(+/-)-8 and lactams (+/-)-9-(+/-)-13, were analysed to evaluate contributions of orthogonal multipolar interactions with the backbone C=O moiety of Asn98 to the free enthalpy of protein-ligand complexation. The lactam derivatives are much more potent and more selective inhibitors (K(i) values between 0.065 and 0.005 microM, selectivity for thrombin over trypsin between 361- and 1609-fold) than the imide compounds (Ki values between 0.057 and 23.7 microM, selectivity for thrombin over trypsin between 3- and 67-fold). Ionic liquid binary mixtures: How different factors contribute to determine their effect on the reactivity

We studied how mixing ionic liquids affected the rate of the Diels Alder reaction between 9-anthracenemethanol and N-ethylmaleimide. The mixtures considered spanned the whole compositional range and differed for the anion or the cation. Synthesis and mesomorphism of related series of triphilic ionic liquid crystals based on 1,2,4-triazolium cations

Abstract The synthesis, liquid crystal and conductivity properties of a series of 27 salts based on the 5-(4-(alkyloxy)phenyl)-1,2,4-triazol-4-ium cation bearing a perfluoroalkyl chain with triflate, tetrafluoroborate and bistriflimide anion are reported. The cations are regarded as triphilic on account of their three distinct regions – hydrocarbon, fluorocarbon and ionic. The anion impact on the self-assembly of naphthalene diimide diimidazolium salts

Naphthalene diimide diimidazolium salts differing in the anion nature were synthesized and their properties as well as their self-assembly behaviour were investigated. In particular, we took into consideration the N,N′-bis-(1-octyl-3-propylimidazolium)-naphthalene diimide cations and anions differing in size, shape and coordination abilities like [I−], [BF4−] and [NTf2−]. Polymides 3a and 3b with stiff-chains bearing trioxaoctyl or tetraoxaundecyl side chains were synthesized and characterized. The polycondensation was done with pyromellitic anhydride and N,N′-bis(1-imidazolylcarbonyl)-2,5-(1,4,7-trioxaoctyl)- or 2,5-(1,4,7,10-tetraoxaundecyl)-1,4-phenylenediamine (2a or 2b). 3b, being sufficiently soluble, was characterized by 13C NMR spectroscopy. The results of IR and 13C NMR spectroscopy prove the presence of the correct imide structure. In solid state 3a and 3b form layered structures with the side chains being in a disordered state.

Differential effects of cysteine and methionine residues in the antioxidant activity of human serum albumin

Antioxidant properties of human serum albumin (HSA) may explain part of its beneficial role in various diseases related to free radical attack. In the present study, the antioxidant role of Cys and Met was studied by copper-mediated oxidation of human low density lipoproteins and by free radical-induced blood hemolysis which essentially assessed metal-chelating and free radical scavenging activities, respectively. Mild conditions were set up to specifically modify Cys and Met residues by N-ethylmaleimide (NEM) and chloramine T treatments, respectively. Succinic derivatives of inulin (INU-SA) with two different degrees of derivatization (20% and 30%, mol/mol) were cross-linked with α,β-polyaspartylhydrazide (PAHy) to obtain INUPAHy hydrogels. Cross-linking was performed using N-ethyl-N-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC) and N-hydroxysulfosuccinimide (NHSS) as coupling agents and by varying the reaction time (4 h, 8 h and 24 h).
