The severe phenotype of Diamond-Blackfan anemia is modulated by heat shock protein 70.

International audience; Diamond-Blackfan anemia (DBA) is a rare congenital bone marrow failure syndrome that exhibits an erythroid-specific phenotype. In at least 70% of cases, DBA is related to a haploinsufficient germ line mutation in a ribosomal protein (RP) gene. Additional cases have been associated with mutations in GATA1. We have previously established that the RPL11+/Mut phenotype is more severe than RPS19+/Mut phenotype because of delayed erythroid differentiation and increased apoptosis of RPL11+/Mut erythroid progenitors. The HSP70 protein is known to protect GATA1, the major erythroid transcription factor, from caspase-3 mediated cleavage during normal erythroid differentiation.…

Viral highway to nucleus exposed by image correlation analyses.

AbstractParvoviral genome translocation from the plasma membrane into the nucleus is a coordinated multistep process mediated by capsid proteins. We used fast confocal microscopy line scan imaging combined with image correlation methods including auto-, pair- and cross-correlation, and number and brightness analysis, to study the parvovirus entry pathway at the single-particle level in living cells. Our results show that the endosome-associated movement of virus particles fluctuates from fast to slow. Protoparvoviruses target the nucleus due to their dependence on the cellular reproduction machinery during the replication and expression of their single-stranded DNA genome. In recent years, our understanding of the multistep process of the capsid nuclear import has improved, and led to the discovery of unique viral nuclear entry strategies. Responsiveness and Minimal Clinically Important Difference of the Five Times Sit-to-Stand Test in Patients with Stroke

This study aimed to analyze the responsiveness of the 5STS test among stroke patients and to estimate the MCIDs (minimal clinically important differences) for different severity levels of community ambulation and stages of recovery. The 5STS and comparator instruments (gait speed and Functional Ambulatory Category (FAC)) were evaluated at baseline. These measures were repeated at 4 (Stage 1) and 8 weeks (Stage 2), together with the Global Rating of Change (GROC). This empirical research explores the perceptions and attitudes of employers in hiring and retaining people with borderline intellectual functioning (BIF). La satisfacció sexual s?ha convertit en una qüestió important dintre dels estudis sobre sexualitats, dels que sistemàticament les persones grans queden excloses al reduir la seua sexualitat a aspectes relacionats amb la mala salut. En aquest estudi s?analitza la satisfacció sexual amb la parella d?aquest grup de població a través de la Encuesta Nacional de Salud Sexual, prenent com a base el model biopsicosocial de la sexualitat (DeLamater, 2002) i considerant el gènere i diferents grups d?edat. Els resultats indiquen el caràcter multifactorial de la satisfacció sexual, prioritzant elements de natura relacional, social i cultural per sobre de l?estat de salut.
