Folate-mediated targeting of polymeric conjugates of gemcitabine.
2005
The synthesis of two new macromolecular prodrugs for active tumor targeting was set up. Gemcitabine (2'-deoxy-2',2'-difluorocytidine) was conjugated to alpha,beta-poly(N-2-hydroxyethyl)-DL-aspartamide (PHEA) through succinyl or diglycolyl hydrolysable spacers. The targeting agent folic acid was attached to the macromolecular backbone through the aminocaproic spacer. The two conjugates [PHEA-(5'-succinylgemcitabine)-1'-carboxypentyl-folamide and PHEA-(5'-diglycolyl-gemcitabine)-1'-carboxypentyl-folamide], were purified and extensively characterised by spectroscopic (UV, IR and NMR) and chromatographic analyses to determine the correct chemical structure, the purity degree and the reaction yi…
Application of Pediococcus acidilactici LUHS29 immobilized in apple pomace matrix for high value wheat-barley sourdough bread
2017
The aim of this study was to evaluate the potential use of Pediococcus acidilactici LUHS29 immobilized in apple pomace in case to apply in barley sourdough fermentation for functional bread production. The strain was phenotypically characterized by the growth and acidification rate, carbohydrate metabolism and resistance to acidic conditions. The effect of immobilized bacterial cells on antioxidant properties of barley sourdough and on the acrylamide content in wheat-barley bread was analyzed. The phenotypic and molecular testing indicates the P. acidilactici having a versatile carbohydrate metabolism and acid resistance, showing 42.7% of viable cells surviving after incubation at low pH as…
A specific microassay for evaluating hepatic LDH activity in co-cultures of hepatocytes with other cells.
1994
This study describes the development of a simple, rapid and reproducible microassay for determining the intracellular LDH activity of rat hepatocytes present in a co-culture system with other cells. The procedure involves treatment of cellular homogenates with an anti-LDH antiserum that specifically inhibits the LDH activity of rat hepatocytes. The assay is performed in 96-well plates and LDH activity can be measured directly in the same wells using a colorimetric method. The difference in LDH activity values measured before and after antiserum incubation reflects the LDH content of the hepatocytes in the sample. The advantages of this method are the small number of cells required, a reduct…
Influence of nitrogen and carbon sources on the production of ochratoxin A by ochratoxigenic strains of Aspergillus spp. isolated from grapes.
2008
This work studies the influence of nitrogen and carbon source on ochratoxin A production by three Aspergillus isolates A. ochraceus (Aso2), A. carbonarius (Ac25) and A. tubingensis (Bo66), all isolated from grapes. A basal medium (0.01 g/l FeSO4.7H2O, 0.5 g/l MgSO4.7H2O, 0.5 g/l Na2HPO4.2H2O, 1.0 g/l KCl) was prepared. This medium was supplemented with different nitrogen sources, both inorganic [(NH4)3PO(4), 0.3 g/l plus NH4NO3, 0.2 g/l] and organic (histidine, proline, arginine, phenylalanine, tryptophan or tyrosine) at two concentrations (0.05 g/l or 0.3 g/l), and different carbon sources (sucrose, glucose, maltose, arabinose or fructose) at three concentrations (10 g/l, 50 g/l or 150 g/l…
Arachnid oenocytes: ecdysone synthesis in the legs of harvestmen (Opilionidae).
1981
Cells measuring up to 130 microns have been found in the proximal segments of the femora of all four pairs of walking legs in various species of harvestmen (Phalangium opilio, Leiobonum limbatum, Opilio parietinus, and Opilio ravennae). These cells exhibit all the fine-structural characteristics of insect oenocytes, in particular the conspicuous agranular endoplasmic reticulum. Radioimmunoassay after in vitro incubation of these cells has demonstrated the synthesis of alpha- and beta-ecdysone. These ecdysteroids have been found in the ovaries and tergites of the opisthosoma as well as in the oenocytes.
Effects of tributyltin(IV) chloride on fertilization of Styela plicata (Ascidiacea: Tunicata): II. Scanning and transmission electron microscopy stud…
2003
The morphological aspects of Styela plicata fertilization after treatment with tributyltin(IV) chloride are described by means of scanning and transmission electron microscopy investigations. Alterations have been shown both on female and male gametes; spermatozoa, all the egg envelopes and the mitochondria of the egg cortical cytoplasm are modified in relation to incubation time. As a consequence, the damage to gametes blocks sperm-egg interaction and fertilization does not occur. Copyright © 2003 John Wiley & Sons, Ltd.
Impacts of ambient temperature and clutch size on incubation behaviour onset in a female-only incubator songbird
2021
Ambient temperature is assumed to be the major cue used by passerines to synchronize their laying and hatching dates to the expected peak of prey availability. While laying eggs, females are still able to fine-tune their hatching date following increasing or decreasing patterns of ambient temperature, mostly via changes in incubation onse t. The onset of incubation behaviour in relation to the laying sequence could have later conse- quences for the duration of the incubation period and the extent of hatching asynchrony. Clutch size is also known to affect incubation patterns and might therefore condition potential responses to changing temperatures. In this study we assessed the association…
Incubation and hatching periods in a Mediterranean Great Tit Parus major population
2014
Capsule The onset of incubation relative to clutch completion is highly variable in Great Tits Parus major, and has important consequences for the duration of the incubation and hatching periods.Aim To investigate when incubation starts relative to clutch completion, its effects on the length of the incubation and hatching periods, and which proximate factors affect all of these traits. Methods We used data from a Great Tit population in Eastern Spain collected over 15 years. Periodic visits to the nests (daily at some stages) allowed the determination of breeding parameters of interest. General linear models were used for analyses. Results On average, incubation started the day of laying of …
Effect of Incubation Body Mass on Reproductive Success and Survival of Two European Diving Ducks: A Test of the Nutrient Limitation Hypothesis
1997
The nutrient limitation hypothesis postulates that variation in nutrient reserves during egg laying and incubation are important predictors of future reproductive success of female waterfowl. We tested one component of this hypothesis in two European diving ducks, Common Pochard (Aythya ferina) and Tufted Duck (Aythya fuligula), relating body masses of incubating females to subsequent nest success, and to duckling and adult survival. We based our analyses on 3,224 Common Pochards and 1,812 Tufted Ducks captured and weighed during the last two weeks before ducklings hatched at Engure Marsh, Latvia, in 1978-1993. Adult (ASY) females from successful nests weighed significantly more than those …
Fluorescence-Based Screening of Cytochrome P450 Activities in Intact Cells
2013
Fluorimetric methods to assess cytochrome P450 (P450) activities that do not require metabolite separation have been developed. These methods make use of non- or low-fluorescent P450 substrates that produce highly fluorescent metabolites in aqueous solutions. The assays are based on the direct incubation of intact cells in culture with appropriate fluorogenic probe substrates, followed by fluorimetric quantification of the product formed and released into incubation medium. We describe a battery of fluorescence assays for rapid measurement of the activity of nine P450s involved in drug metabolism. For each individual P450 activity the probe showing the best properties (highest metabolic rat…
