We have checked the ability of the Candida albicans GAPDH polypeptide, which lacks a conventional N-terminal signal peptide, to reach the cell wall in Saccharomyces cerevisiae by using an intracellular form of the yeast invertase as a reporter protein. The grapevine downy mildew affects leaves and young berries and can affect the harvest quality and quantity. The causal agent is the obligate biotroph oomycete Plasmopara viticola that induces severe physiological alterations in infected leaves. One is the abnormal accumulation of starch in oil spots, the characteristic symptoms of the disease. Another is a deregulation of stomatal movements. Activities of NAD(+)-dependent sorbitol dehydrogenase (SDH), sorbitol oxidase (SOX), sucrose synthase (SS), acid invertase (AI), and neutral invertase (NI) in 'Encore' peach (Prunus persica L.) fruits and developing shoot tips were assayed during the growing season to determine whether carbohydrate metabolizing enzymes could serve as indicators of sink strength. Separation of chitosomes and secretory vesicles from the ?slime? variant of Neurospora crassa

Cells from the “slime” variant of Neurospora crassa were broken in isotonic conditions by use of triethanolamine buffer plus EDTA. After removal of large membranous structures by low-speed centrifugation, chitosomes and secretory vesicles were separated by means of gel filtration, precipitation of membranous contaminants with Concanavalin A, and centrifugation in sucrose or glycerol gradients. Polypeptidic composition of fractions enriched in secretory vesicles or chitosomes was found to be distinct.
