Rapid antibiotic susceptibility testing on blood cultures using MALDI-TOF MS

International audience; Antibiotic resistance is a major public health problem requiring the early optimization of antibiotic prescriptions. Matrix-Assisted Laser Desorption Ionization-Time Of Flight Mass Spectrometry (MALDI-TOF MS) has been shown to accurately identify bacteria from positive blood culture. Here, we developed a rapid detection of Escherichia coli resistance to amoxicillin (AMX) and cefotaxime (CTX) from positive blood culture based on MALDI-TOF MS. The self-assembling of sodium bis (2-ethylhexyl) sulfosuccinate (AOT) in gas phase has been investigated by electrospray ionization- and matrix-assisted laser desorption/ionization mass spectrometry. Large surfactant clusters with an aggregation number close to that found in apolar media have been observed either as positive or negative ions. Moreover, the marked predominance of singly charged species as well as preliminary theoretical calculations strongly suggest an aggregate structure characterized by an internal hydrophilic core hosting the extra charge surrounded by an apolar shell constituted by the surfactant alkyl chains. Classification of vegetable oils according to their botanical origin using amino acid profiles established by direct infusion mass spectrometry.

Amino acid profiles, established by direct infusion mass spectrometry, have been used to classify vegetable oils according to their botanical origin. The proteins present in hazelnut, sunflower, corn, soybean, olive, avocado, peanut and grapeseed oils were precipitated with acetone, and the residue was hydrolyzed in acid medium, diluted in a hydrochloric acid/ethanol mixture, and infused into the mass spectrometer. Determination of alcohols in essential oils by liquid chromatography with ultraviolet detection after chromogenic derivatization

Abstract An HPLC-UV method to determine compounds having a hydroxyl functional group in plant essential oils is developed. The sample is diluted with 1,4-dioxane and the analytes are derivatized with phthalic anhydride. The derivatives (phthalates hemiesters) are separated on a C8 column using an acetonitrile (ACN)/water gradient. Separation conditions were optimized using the DryLab® method development software. For the alcohols and phenols present in mint and rose essential oils, optimization led to a ca. 40 min gradient time and a column temperature of 8 °C. Interactions of Ultra-cold Alkaline-earth-like and Alkali Atoms with Light

This paper presents the experimental results of photoionization and photoassociation experiments performed with both ultra-cold alkaline-earth-like and alkali atoms. Photoionization cross-section was studied for 88Sr at the blue magic 390 nm wavelength as well as for 87Rb at 254 nm. We also present preliminary results of photoassociation spectroscopy of the molecular line detected near the dissociation threshold of the excited Rb*Hg molecule at D1 Rb line (795 nm).

Supramolecular Aggregates in Vacuum: Positively Mono-Charged Sodium Alkanesulfonate Clusters

The formation and structural features of positively mono-charged aggregates of sodium bis(2-ethylhexyl) sulfosuccinate (AOT) and sodium methane—(MetS), butane—(ButS) and octane—(OctS) sulfonate molecules in the gas phase have been investigated by electrospray ionization mass spectrometry, energy-resolved mass spectrometry and density functional theory (DFT) calculations. The experimental results show that the center-of-mass collision energy required to dissociate 50% of these mono-charged aggregates scantly depends on the length of the alkyl chain as well as on the aggregation number. Optical measurements of ground-state properties of short-lived nuclei in resonance cells

Hyperfine structure and isotope shift of radioactive Hg, Au and Cd isotopes have been determined by optical spectroscopy. In all cases the atoms were confined in a resonance cell. The various variants of the cell technique and alternative methods as the resonance ionization spectroscopy related to the investigation of short-lived nuclei are described, and some results are discussed.

The first adiabatic ionization energy and the first singlet–triplet splitting of the amidogen radical (NH2) have been determined by high-level ab initio quantum chemistry based on the coupled-cluster approach (90 041 and 10 319 cm−1, respectively) and by high-resolution pulsed-field-ionization zero-kinetic-energy (PFI-ZEKE) photoelectron spectroscopy (90 083.8 ± 1.0 and 10 222.0 ± 1.3 cm−1, respectively). A comparison between the theoretical and experimental values demonstrates the predictive powers of high-level ab initio theory in the derivation of the thermochemical properties of small molecular systems. AbstractThe ammonium ion binding affinities of tetraethyl resorcarene (1) and its per-methylated derivative (2) were studied by electrospray ionization Fourier transform ion cyclotron resonance (ESI-FTICR) mass spectrometry. Ten different ammonium ions were tested as guests for the resorcarenes. A strong tendency for complex formation was observed with all ammonium ions of size and charge distribution suitable for noncovalent interactions with the cavities of the resorcarene hosts 1 and 2.
