Human dental pulp cell apoptosis: immunohistochemical study after applying orthodontic traction.
2012
The aim of this study was to compare human dental pulp stress and programmed cell death after 3 and 6 months of orthodontic treatments by assessing the degree of apoptosis and related proteins. Human dental pulps were collected from twenty young patients orthodontically treated by Straight Wire technique. Samples were fixed, paraffin-embedded and processed for histology and immunohistochemistry using anti-heat shock protein 60 kDa (Hsp60), -caspase 3, -caspase 9, and -PCNA antibodies, as well as TUNEL reactions. Moreover, we performed immunoprecipitation for Hsp60 and caspase 3, and for Hsp60 and caspase 9, from paraffin extracted tissues. Increased levels of both caspases and Hsp60 occurre…
Matrix metalloproteinase-2 matrix metalloproteinase-9 and inducible nitric oxide synthase in oral leukoplakia: Immunohistochemistry and RT-PCR analys…
2008
Oral leukoplakia is the most common and potentially malignant disorder of the oral mucosa. The definition of leukoplakia given by the World Health Organization is "a white plaque that cannot be characterized either from a clinical or from a histopathological point of view", thus the diagnosis of leukoplakia is based on the exclusion of other lesions of the oral mucosa. We believe it is necessary to identify molecular and immunohistochemical parameters that can contribute to discriminating between the different leukoplakia clinical subtypes coded by the epidemiology. In the present work we show the preliminary results of this research project. We investigated the expression of matrix metallo…
Hsp27 and Hsp60 in human submandibular salivary gland: Quantitative patterns in healthy and cancerous tissues with potential implications for differe…
2021
Tumors of the submandibular salivary gland (SMG) are uncommon but sufficiently frequent for the physician to consider them in routine examinations and for the pathologist to be prepared to differentiate them from other tissue abnormalities. However, scarcity of specimens makes training difficult, a situation compounded by the lack of accepted universal diagnostic guidelines. Furthermore, there is little information on the chaperone system (CS) of the gland, despite the increasing evidence of its participation in carcinogenesis as a biomarker for diagnosis and patient follow up, and in the mechanisms by which the tumor cells thrive. We are investigating this aspect of various tumors, and her…
Human umbilical cord expresses several vasoactive peptides involved in the local regulation of vascular tone: protein and gene expression of Orphanin…
2011
Full-term human umbilical cord contains three blood vessels: two arteries coiled around a vein and surrounded by Wharton’s jelly, a mucous tissue with few mesenchymal stromal cells and abundant extracellular matrix. Umbilical vessels lack innervations, thus endothelial cells must play a role in the control of blood flow. The aim of this study was to investigate in human umbilical cord the expression of five peptides that could be involved in the regulation of vascular tone: Orphanin FQ, Oxytocin, Atrial Natriuretic Peptide (ANP), endothelial Nitric Oxide Synthase (eNOS) and inducible Nitric Oxide Synthase (iNOS). The expression of these molecules in full-term human umbilical cord …
Immunohistochemical and Biomolecular Identification of Orphanin FQ, eNOS, Atrial natriuretic Factor and Oxytocin in Rat Seminal Vesicles
2009
In previous studies performed on rodents, we detected the presence of adreno-cholinergic and peptidergic innervation in seminal vesicles and other organs of the male genital system, such as prostate and ductus deferens, in which we also investigated the expression of NOS and NADPH-diaphorase. During the present project we focused our attention on the expression of some peptides involved in local control of smooth muscle relaxation, contractility, vasodilatation and control of blood flow in rat seminal vesicles. We investigated, through immunohistochemistry and RT-PCR, the presence of four peptides: orphanin, eNOS, ANF and oxytocin. Immunohistochemistry was used to detect the presence of the…
Expression of matrix metalloproteinases in full-term human umbilical cord and Human Umbilical Vein Endothelial Cells.
2010
Background. Matrix metalloproteinases (MMPs) are extracellular zinc-dependent endopeptidases involved in breakdown and remodelling of extracellular matrix in physiological and pathological processes. MMPs have also a role on cell proliferation, migration, differentiation, angiogenesis and apoptosis. Umbilical cord is a special organ subjected to many changes during pre-natal life and whose cells can maintain a certain degree of plasticity also in post-natal period; for example recently they have been used as a source of stem cells. For these reasons we believe it is interesting to investigate the expression of some remodelling enzymes in this tissue. Human Umbilical Vein Endothelial cells (…
Comportamento della eNOS e dell'iNOS nella genesi e mantwnimento delle ulcere venose degli arti inferiori
2019
Chronic venous insufficiency (CVI) and venous ulcers in the lower areas are widespread problems with a prevalence of 1% - 2% in the western population. The pathophysiology of this pathological manifestation is not yet fully clarified and it can be assumed that a multifactorial genesis should be considered. Certainly the high pressure values in the peripheral venous system, the defective valves in the capillary network, the increase in permeability and capillary neogenesis (discharge of liquids in the extracellular compartment - + edema), the discharge of fibrinogen and the subsequent formation of a pericapillary fibrin cuff are the most important mechanisms related to hypoxia and skin isc…
Immunohistochemical Expression of NO-synthase Constitutive Isoform (cNOS)in Mammals Normal Prostate and in Human Prostatic Obstructive Hyperplasia
2007
The aim of present study is to investigate the immunohistochemical identification of cNOS-containing prostatic structures in various species and in hyperplastic human prostate. We used specimens of rat, guinea pig and hamster. Human prostatic specimens were removed from patients suffering of prostatic hyperplasia. The study was processed by ABC method. Immunohistochemical staining with anti nNOS antibody revealed the existence of strong reactivity in numerous ganglionic cell bodies and varicose nerve fibers in the fibromuscular stroma, in smooth cells around acini and in blood vessels endothelium. Surprisingly nNOS immunoreactivity appears in the glandular epithelium. In the hyperplastic pr…
