ALS-linked FUS mutations confer loss and gain of function in the nucleus by promoting excessive formation of dysfunctional paraspeckles

Mutations in the FUS gene cause amyotrophic lateral sclerosis (ALS-FUS). Mutant FUS is known to confer cytoplasmic gain of function but its effects in the nucleus are less understood. FUS is an essential component of paraspeckles, subnuclear bodies assembled on a lncRNA NEAT1. Paraspeckles may play a protective role specifically in degenerating spinal motor neurons. However it is still unknown how endogenous levels of mutant FUS would affect NEAT1/paraspeckles. Using novel cell lines with the FUS gene modified by CRISPR/Cas9 and human patient fibroblasts, we found that endogenous levels of mutant FUS cause accumulation of NEAT1 isoforms and paraspeckles. The use of centre-pivot irrigation systems has significantly increased in the last few decades, since they are easier and less time-consuming to manage compared to other irrigation systems. Standard high-pressure impact sprinklers, low-pressure spray sprinklers or LEPA (Low Energy Precision Application) systems are usually combined with a centre-pivot irrigation system (CP-SYS). International audience; Aging is frequently associated with a decreased postural stability, essentially after 60 years, leading to an increased risk of falling. In this article we propose to highlight the influence of the aging of the neuromuscular system on postural stability when standing upright. To maintain balance while standing upright, human needs to control the activity of ankle muscles and particularly the plantar flexors. Recurrent hypoglycemia (RH) impairs the counter-regulatory response (CRR) which restores euglycemia. During hypoglycemia, ventromedial hypothalamus (VMH) production of nitric oxide (NO) and activation of its receptor soluble guanylyl cyclase (sGC) are critical for the CRR. Theoretical study on travelling web dynamics and instability under non-homogeneous tension

Problems of dynamics and stability of a moving web, travelling between two rollers at a constant velocity, are studied using analytical approaches. Transverse vibrations of the web are described by a partial differential equation that includes the centrifugal force, in-plane tension, elastic reaction and nonstationary inertial terms. The model of a thin elastic plate subjected to bending and non-homogeneous tension is used to describe the bending moment and the distribution of membrane forces. Static instability analysis for travelling membranes and plates interacting with axially moving ideal fluid

The out-of-plane instability of a moving plate, travelling between two rollers with constant velocity, is studied, taking into account the mutual interaction between the buckled plate and the surrounding, axially flowing ideal fluid. Transverse displacement of the buckled plate (assumed cylindrical) is described by an integro-differential equation that includes the centrifugal force, the aerodynamic reaction of the external medium, the vertical projection of membrane tension, and the bending force. The objective of the present study was to evaluate the parameters, which characterize the pulsing flow of the gas and liquid through a bed, namely the frequency of pulsation, the velocity of the pulses and the pulse structure, for foaming systems. The paper presents the results of experiments aimed at determining the effect of the foaming power and the surface tension of liquid phase on the values of the measured parameters.
