Use of deep learning methods to translate drug-induced gene expression changes from rat to human primary hepatocytes
2020
In clinical trials, animal and cell line models are often used to evaluate the potential toxic effects of a novel compound or candidate drug before progressing to human trials. However, relating the results of animal and in vitro model exposures to relevant clinical outcomes in the human in vivo system still proves challenging, relying on often putative orthologs. In recent years, multiple studies have demonstrated that the repeated dose rodent bioassay, the current gold standard in the field, lacks sufficient sensitivity and specificity in predicting toxic effects of pharmaceuticals in humans. In this study, we evaluate the potential of deep learning techniques to translate the pattern of …
Genetic counselling difficulties and ethical implications of incidental findings from array-CGH: a 7-year national survey
2016
Microarray-based comparative genomic hybridization (aCGH) is commonly used in diagnosing patients with intellectual disability (ID) with or without congenital malformation. Because aCGH interrogates with the whole genome, there is a risk of being confronted with incidental findings (IF). In order to anticipate the ethical issues of IF with the generalization of new genome-wide analysis technologies, we questioned French clinicians and cytogeneticists about the situations they have faced regarding IF from aCGH. Sixty-five IF were reported. Forty corresponded to autosomal dominant diseases with incomplete penetrance, 7 to autosomal dominant diseases with complete penetrance, 14 to X-linked di…
Frailty Quantified by the "Valencia Score" as a Potential Predictor of Lifespan in Mice.
2017
The development of frailty scores suitable for mice and which resemble those used in the clinical scenario is of great importance to understand human frailty. The aim of the study was to determine an individual frailty score for each mouse at different ages and analyze the association between the frailty score and its lifespan. For this purpose, the "Valencia Score" for frailty was used. Thus, a longitudinal study in mice was performed analyzing weight loss, running time and speed, grip strength and motor coordination at the late-adult, mature and old ages (40, 56 and 80 weeks old, respectively). These parameters are equivalent to unintentional weight loss, poor endurance, slowness, weaknes…

Increased survival of honeybees in the laboratory after simultaneous exposure to low doses of pesticides and bacteria
2018
Recent studies of honeybees and bumblebees have examined combinatory effects of different stressors, as insect pollinators are naturally exposed to multiple stressors. At the same time the potential influences of simultaneously occurring agricultural agents on insect pollinator health remain largely unknown. Due to different farming methods, and the drift of applied agents and manure, pollinators are most probably exposed to insecticides but also bacteria from organic fertilizers at the same time. We orally exposed honeybee workers to sub-lethal doses of the insecticide thiacloprid and two strains of the bacterium Enterococcus faecalis, which can occur in manure from farming animals. Our re…
Facilitation of Health Professionals Responsible Autonomy with Easy-to-Use Hospital Data Querying Language
2018
Support for the development of responsible autonomy as opposite to management that is based on direct control is found to be by far more effective approach in healthcare management, especially when it concerns physicians as the most influential group of health professionals. It is therefore important to obtain a process-oriented knowledge system where physicians would be able to autonomously answer questions which are outside the scope of pre-made direct control reports. However, the ad-hoc data querying process is slow and error-prone due to inability of health professionals to access data directly without involving IT experts. The problem lies in the complexity of means used to query data…
Spontaneous brain processing of the mammary pheromone in rabbit neonates prior to milk intake.
2016
International audience; Chemical signals play a critical role in interindividual communication, including mother-young relationships. Detecting odor cues released by the mammary area is vital to the newborn's survival. European rabbit females secret a mammary pheromone (MP) in their milk, which releases sucking related orocephalic movements in newborns. Pups spontaneously display these typical movements at birth, independently of any perinatal learning. Our previous Fos mapping study (Charra et al., 2012) performed in 4-day-old rabbits showed that the MP activated a network of brain regions involved in osmoregulation, odor processing and arousal in comparison with a control odor. However, a…
Changing the education paradigm in oncology: ESO masterclass, 17 years of continuous success
2020
In this review, we summarize the history of the 41 Masterclasses in Clinical Oncology (MCO) organized by ESO or ESO-ESMO during the last 17 years. MCOs have been held in five different geographical regions including: a) Central Europe, b) Eastern Europe and Balkans, c) Baltic and Euroasia, d) Arab World and Southern European Countries and e) Latin America. More than 2.000 young oncologists have attended and more than 250 distinguished faculty members have actively participated. The program exposes students to sessions covering all major tumors ("big killers") and to spotlights updating information on various important cancers and related topics. Participants are able to present their own cl…
PSPU-Net for Automatic Short Axis Cine MRI Segmentation of Left and Right Ventricles
2020
[EN] Characterization of the heart anatomy and function is mostly done with magnetic resonance image cine series. To achieve a correct characterization, the volume of the right and left ventricle need to be segmented, which is a timeconsuming task. We propose a new convolutional neural network architecture that combines U-net with PSP modules (PSPU-net) for the segmentation of left and right ventricle cavities and left ventricle myocardium in the diastolic frame of short-axis cine MRI images and compare its results against a classic 3D U-net architecture. We used a dataset containing 399 cases in total. The results showed higher quality results in both segmentation and final volume estimati…
Deep learning for diagnosis and survival prediction in soft tissue sarcoma.
2021
Background Clinical management of soft tissue sarcoma (STS) is particularly challenging. Here, we used digital pathology and deep learning (DL) for diagnosis and prognosis prediction of STS. Patients and methods Our retrospective, multicenter study included a total of 506 histopathological slides from 291 patients with STS. The Cancer Genome Atlas cohort (240 patients) served as training and validation set. A second, multicenter cohort (51 patients) served as an additional test set. The use of the DL model (DLM) as a clinical decision support system was evaluated by nine pathologists with different levels of expertise. For prognosis prediction, 139 slides from 85 patients with leiomyosarcom…
