Ultrahigh-Throughput Proteomics Using Fast RPLC Separations with ESI-MS/MS

We describe approaches for proteomics analysis using electrospray ionization-tandem mass spectrometry coupled with fast reversed-phase liquid chromatography (RPLC) separations. The RPLC separations used 50-microm-i.d. fused-silica capillaries packed with submicrometer-sized C18-bonded porous silica particles and achieved peak capacities of 130-420 for analytes from proteome tryptic digests. Solution and thermal behaviour of novel dicationic imidazolium ionic liquids

A new class of functionalised dicationic ionic liquids, containing a central cationic unit capped by a basic functionality (imidazole), has been synthesised. These salts have been characterised in isotropic solution using proton and 2D-NMR spectroscopy, and their thermal stability has been studied by DSC and TGA. All these novel salts contain the 1-(1-imidazolylmethyl)-3,5-di{1-(3'-octylimidazolylmethyl)}-benzene cation as a defining structural motif. Ohmic drop of Prussian-blue/graphite+epoxy electrodes

Abstract Ohmic drop effects on the voltammogram shape of the Prussian-blue⇌Everitt’s salt system have been studied by considering the electroactive film model. As the uncompensated resistance varies, the experimental characteristic curve parameters also varies. A dependence of the voltammograms with the graphite proportion was found related with the ohmic drop due to the composite electrodes. The optimal graphite proportion to obtain a good faradaic-to-charge current ratio was 62% in weight which coincides with the second percolation threshold. The ohmic overpotential, which relates the ohmic drop with the electrochemical magnitudes, is also minimum at this proportion.

Chronoamperometry of prussian blue films on ITO electrodes: Ohmic drop and film thickness effect

Abstract The chronoamperograms associated with the reduction of prussian blue films deposited onto indium tin oxide (ITO) electrodes to the Everitt’s salt form, are influenced by the ohmic drop effect. These chronoamperometric curves have been simulated by means of a numerical finite difference model which is able to explain their shape and their dependence on the thickness of the film and on the uncompensated resistance. An analytical expression which describes the dependence of current against time at initial times considering the ohmic drop effect has also been proved when applied to these chronoamperometric curves at short times.

Physicochemical compatibility and stability of nebulizable drug admixtures containing Dornase alfa and tobramycin.

The objective of this in-vitro study was to determine whether admixtures of the inhalation solutions Pulmozyme(®) (Dornase alfa) and either Bramitob(®) or Tobi(®) (both containing Tobramycin) are physicochemically compatible and to analyze the aerodynamic parameters of these admixtures. After mixing, test solutions were stored at room temperature and under ambient light conditions over a period of 24 h. Tobramycin concentrations were determined by using a fluorescence immunoassay. Differential changes in purine nucleotides after Doxorubicin treatment of human cancer cells in vitro

The present investigation was performed to elucidate the role of purine nucleotides as potential indicators of chemosensitivity of malignant tumors. Drug-sensitive (s) and -resistant (r) tumor cell lines grown as monolayers (s: T47D, MCF-7 wild-type; r: NCI/ADR-RES, MCF-7/MDR) or as multicellular spheroids (T47D; NCI/ADR-RES) were exposed to 0.1, 1.0, and 10.0 microM Doxorubicin for up to 24 h. Purine nucleotides were assayed using HPLC and with some selected spheroids using imaging bioluminescence.
