A single mutation in the recombinant light chain of tetanus toxin abolishes its proteolytic activity and removes the toxicity seen after reconstituti…

1994

Specific proteolysis by the tetanus toxin light chain of a vesicle-associated membrane protein (VAMP) involved in exocytosis is thought to underlie its intracellular blockade of neurotransmitter release. To substantiate this mechanism, recombinant light chain was expressed as a maltose binding protein-light chain fusion product in Escherichia coli. After purification of affinity chromatography and cleavage with factor Xa, the resultant light chain was isolated and its identity confirmed by Western blotting and N-terminal sequencing. It exhibited activity similar to that of the native light chain in proteolyzing its target in isolated bovine small synaptic vesicles and in hydrolyzing a 62-re…

Chemical composition and microencapsulation suitability of sumac (Rhus coriaria L.) fruit extract

2021

Sumac (Rhus coriaria L.), a spice obtained by grinding whole sumac berries, is a complex natural product with a plethora of biological activities that can be favorably explored in nutraceutical and pharmaceutical fields. Sicilian sumac is herein chemically investigated by means of a combined NMR/HR MS-based profiling. A hydroalcoholic extract was prepared and its complexity unraveled following fractionation in an alcoholic and an aqueous fraction. The 1H-NMR spectrum of this latter fraction was dominated by the signals of gallic acid and its derivatives, whereas HR MS and HR MS/MS analyses highlighted the diversity in malic acid derivatives. Gallotannins and galloyl flavonol glycosides with…

Phytochemical investigation of the needles of Abies nebrodensis (Lojac.) Mattei

2019

Abies nebrodensis (Lojac.) Mattei (Pinaceae) is a species living in a very small population only in a confined area of Sicily. In this study, the dichloromethane extract of the leaves was analyzed. Apart from three already known metabolites namely dehydroabietic acid; maltol; and rheosmin, previously detected in other species of Abies, a lanostane derivative was isolated. Its chemical structure was elucidated by means of extensive spectroscopic methods.

Chondrichthyans from the Spanish Triassic (westernmost Tethys) with remarks on the evolution of enameloid

2020

The Triassic sediments of the Iberian Peninsula has been the focus of Geological and Paleontological studies for more than a century and the stratigraphic sequence and the invertebrate assemblages are well established for a number of different basin. However, works focused on chondrichthyans faunas were nonexistent until recently. As a consequence, it has been long time assumed that chondrichthyans were very scarce (or inexistent) in the coastal environments of the current Iberian Peninsula during Triassic times. This view changed at the beginning of the last decade, with the studies of paleontologist from the department of Geology of the University of Valencia (Dra. Márquez-Aliaga, Dr. Héc…
