Production of sulfur flavors by ten strains of Geotrichum candidum

ABSTRACT Ten strains of Geotrichum candidum were studied on a liquid cheese model medium for the production of sulfur compounds which contribute to the aroma of cheeses. The volatile components produced by each cultured strain were extracted by dynamic headspace extractions, separated and quantified by gas chromatography (GC), and identified by GC-mass spectrometry. It was shown that four strains of this microorganism produced significant quantities of S -methyl thioacetate, S -methyl thiopropionate, S -methyl thiobutanoate, S -methyl thio iso butanoate, S -methyl thio iso valerate, and S -methyl thiohexanoate. A method for the isolation of xanthomegnin, viomellein, rubrosulphin, viopurpurin, and brevianamide A from Penicillium viridicatum (DSM 2447) is described. After extraction, HPLC was performed with a preparative silicagel column, eluted with toluene / ethyl acetate / formic acid (27/9/1, v/v/v) and dichloromethane / acetic acid (9/1, v/v). The toxins were detected with a UV-monitor. It was possible to isolate them in an absolutely pure state. The described method is operationally simple and very efficient.

Graphitized Carbon Black (GCB) extractive cartridges are evaluated for on-line coupling with a C8 analytical column to determine eleven carbamates and one carboximide pesticide from spiked deionized water at the 1.2 μg/L level. Several experiments were carried out to ascertain whether GCB saturation, pesticide degradation on the surface, existence of by-pass channels, mobility among the bulk cartridge, or strong retention on the surface interfere with the determination of pesticides. Degradation products of chlorsulfuron, chlortoluron, diuron, fluometuron, isoproturon, linuron, metabenzthiazuron, metobromuron, and monuron formed in the gas chromatographic injector have been used for identification of the respective herbicides. Mass spectra of the derived compounds were obtained with a quadrupole mass spectrometric detector working in scan mode (20–450 amu). Reversed-phase packings were synthesized by reaction of n-alkyldimethylchloro- and trifluoroacetoxysilanes with silicas of 15 and 25 nm pore size, employing tetrahydrofuran, N,N-dimethylformamide, benzene, diethyl ether, dichloromethane and acetonitrile as solvents and triethylamine, pyridine, triazole, quinuclidine, 2,4- and 2,6-lutidine and imidazole as bases. To improve the quality level of plastic packaging materials, we determined the influence of chemical degradation of polypropylene (PP) upon the amount of volatile components extracted by hot water from PP pellets. Two formulations were analyzed and compared: PP CR (degraded) and PP (not degraded). After Likens-Nickerson's extraction of the pellets and concentration of the extract, an HPLC fractionation was made to collect three fractions of different poiarities: a pentane (A), a dichloromethane (B), and an ether (C) fraction. The thin-layer chromatography of chlorinated anisoles (methoxybenzenes) and veratroles (1,2-dimethoxybenzenes) has been examined on silica gel G60 and RP-18 thin-layer plates. More than fifty solvent systems were screened and some of them recommended for particular separations. Acetone was shown to be suitable for group separation of both chlorinated anisoles and veratroles on a silica gel G60 stationary phase having a very narrow range of RF values. On the other hand, benzene, dichloromethane and the various mixtures of light petroleum (b.p. 40–60°C) and a more polar eluent (such as diethyl ether, acetone or ethyl acetate) were recommended for separation of certain individual isomers. Isobaric vapor−liquid equilibria for the binary system chloroform + methanol as well as the vapor−liquid equilibria for the chloroform + methanol + [emim][triflate] ternary system have been obtained at 100 kPa using a recirculating still. The ternary ionic liquid (IL) system presents an immiscibility zone at high chloroform composition, which increases with the temperature. Liquid−liquid equilibria for this system have also been determined.
