In silico characterization of the neural alpha tubulin gene promoter of the sea urchin embryo Paracentrotus lividus by phylogenetic footprinting

During Paracentrotus lividus sea urchin embryo development one alpha and one beta tubulin genes are expressed specifically in the neural cells and they are early end output of the gene regulatory network that specifies the neural commitment. In this paper we have used a comparative genomics approach to identify con- served regulatory elements in the P. lividus neural alpha tubulin gene. To this purpose, we have first isolated a genomic clone containing the entire gene plus 4.5 Kb of 5 0 upstream sequences. Ectopic hbox12 Expression Evoked by Histone Deacetylase Inhibition Disrupts Axial Specification of the Sea Urchin Embryo

Dorsal/ventral patterning of the sea urchin embryo depends upon the establishment of a Nodal-expressing ventral organizer. Recently, we showed that spatial positioning of this organizer relies on the dorsal-specific transcription of the Hbox12 repressor. Building on these findings, we determined the influence of the epigenetic milieu on the expression of hbox12 and nodal genes. We find that Trichostatin-A, a potent and selective histone-deacetylases inhibitor, induces histone hyperacetylation in hbox12 chromatin, evoking broad ectopic expression of the gene. Early asymmetric cues triggering the dorsal/ventral gene regulatory network of the sea urchin embryo

Dorsal/ventral (DV) patterning of the sea urchin embryo relies on a ventrally-localized organizer expressing Nodal, a pivotal regulator of the DV gene regulatory network. However, the inceptive mechanisms imposing the symmetry-breaking are incompletely understood. In Paracentrotus lividus, the Hbox12 homeodomain-containing repressor is expressed by prospective dorsal cells, spatially facing and preceding the onset of nodal transcription. Uso delle eparine a basso peso molecolare in chirurgia generale e d'urgenza

L’Eparina è un mucopolisaccaride estratto da alcuni organi quali il polmone, fegato e mucosa intestinale e si lega alla superficie delle cellule endoteliali. La sua attività biologica richiede la presenza di un cofattore plasmatico ad attività inibitrice proteasica [Antitrombina III], che inibisce i fattori proteasici della coagulazione formando con essi complessi stabili equimolari. In assenza di eparina, tali reazioni sono lente, mentre al contrario sono accelerate di quasi 1000 volte. Several studies showed that Fetuin-a is an extracellular calcium-regulatory protein acting as a potent inhibitor of calcium-phosphate precipitation, involved also in bone metabolism because of its high affinity for hydroxyapatite. The aim of this study is to evaluate the serum Fetuin-A levels in patients with chronic renal failure before and after a single session of dialysis and to investigate the correlation between protein serum concentrations and T256S genotype. Fetuin-A in elderly: effect of genotype on serum levels

Fetuin-A is a circulating inhibitor of ectopic calcification in vivo; it is decreased in patients with chronic kidney disease and correlates with glomerular filtration rate in patients with coronary artery disease. However, serum levels of Fetuin-A has not been described in elderly. Aim of the study was to evaluate Fetuin-A serum levels together with AHSG T256S genotype in a population of healthy elderly. Serum fetuin-A levels were determined by ELISA. T256S polymorphism of AHSG gene was determined by PCR-RFLP. Serum Fetuin-A was 0,38 ± 0,13 g/l in the group composed by 206 healthy centenarians.
