Molecular weights of Trogamid T® were determined by ultracentrifugation, light-scattering, osmometry and viscometry, molecular weight distributions by gel permeation chromatography (GPC) and five different fractionation methods. The separation efficiency of GPC with N,N-dimethylformamide as solvent was tested with three different types of gels, crosslinked polystyrene, porous glass and crosslinked poly(vinyl acetate), the last one yielding the best results. Based on the highly disparate reactivities of isoprene (I, rI = 25.4) and 4-methylstyrene (4MS, r4MS = 0.007) in the anionic copolymerization in nonpolar media, a general strategy for the rapid and scalable synthesis of tapered multiblock copolymers with an extremely steep gradient has been developed. A repetitive addition strategy of a mixture of isoprene and 4MS leads to a tapered diblock in each case, giving access to linear alternating multiblock copolymers of the (AB)n type with up to 10 blocks. The transfer of the food-borne pathogen Listeria monocytogenes from 30 to 5 degrees C was characterized by the sharp induction of a low molecular mass protein. This major cold shock protein has an isoelectric point at pH 5.1 and a molecular mass of about 18 kDa, as observed on two-dimensional gel electrophoresis (2-DE) pattern. Its N-terminal sequence, obtained from the 2-DE spot, shared a complete sequence identity with a Listeria innocua non-heme iron-binding ferritin. Keyhole limpet hemocyanin (KLH) is a mixture of two hemocyanin isoforms, termed KLH1 and KLH2. Within KLH1 eight oxygen-binding functional units (FUs), 1-a to 1-h, have been identified, in contrast to KLH2, which was previously thought to be organized in seven FUs (2-a to 2-g). By limited proteolysis of KLH2 subunits, isolation of the polypeptide fragments, and N-terminal sequencing, we have now identified an eighth FU of type h, with a molecular mass of 43 kDa. Treatment with heavy metals, such as nickel, lead or cadmium, elicits different cellular stress responses according to the metal used and the length of treatment. In Paracentrotus lividus embryos the inducible forms of HSP70 (HSP70/72) are different in molecular mass from the constitutively expressed HSP75, and they can be used as markers of cellular stress. Even a short treatment with each metal induces the synthesis of HSP70/72 which remain stable for at least 20 h and differ little in their isoelectric points. A monoclonal antibody raised against monkey liver phenylalanine hydroxylase (PAH) has been used to detect this protein in Drosophila melanogaster. A cross-reacting material (CRM) band of apparent molecular mass 50-52 kDa, equivalent to that deduced for the Drosophila melanogaster PAH protein based on the pah gene cDNA sequence, has been detected. This CRM was analysed throughout development and showed an equivalent pattern to that reported for PAH activity in this insect, with maxima at pupariation and at pharate adult formation.
