Combined Carboplatin plus Ifosfamide and Cisplatin in Patients with Advanced Ovarian Carcinoma. A Phase I–II Study
1998
Abstract Because of the relative lack of overlapping toxicity, carboplatin (PPL) and cisplatin (CDDP) can be easily combined for treatment of ovarian cancer to increase total platinum dose intensity. Ifosfamide (IFO), one of the most effective single agents in ovarian cancer, has a low hematological toxicity when administered in continuous infusion. From January 1991 to December 1993, 34 patients with advanced ovarian cancer, previously untreated with chemo- or radiotherapy, were enrolled in a phase I–II study with the aim of determining the maximum tolerated dose (MTD) of CDDP (on day 8 of a 28-day cycle) in combination with PPL (300 mg/m 2 on day 1) and IFO (4,000 mg/m 2 /24 h by continuo…
Alkali consumption of aliphatic carboxylic acids during alkaline pulping of wood and nonwood feedstocks
2013
Abstract The carbohydrate degradation products have been examined, which are formed during the conventional kraft pulping of a softwood, hardwoods, bamboo, and wheat straw as well as soda and soda-anthraquinone pulping of wheat straw. The focus was on “volatile” acids such as formic and acetic acids and “nonvolatile” hydroxy monocarboxylic and dicarboxylic acids. The different consumption profiles were obtained for the charged alkali required for the neutralization of these aliphatic acids depending on the feedstock and the cooking method. The relative composition of the acid fraction in the black liquors of softwood and hardwood and nonwood feedstocks showed characteristic variations. Howe…

Black Hole Shadow Drift and Photon Ring Frequency Drift
2021
The apparent angular size of the shadow of a black hole in an expanding Universe is redshift-dependent. Since cosmological redshifts change with time - known as the redshift drift - all redshift-dependent quantities acquire a time-dependence, and a fortiori so do black hole shadows. We find a mathematical description of the black hole shadow drift and show that the amplitude of this effect is of order $10^{-16}$ per day for M87$^{\star}$. While this effect is small, we argue that its non-detection can be used to constrain the accretion rate around supermassive black holes, as well as a novel probe of the equivalence principle. If general relativity is assumed, we infer from the data obtaine…
Shining primordial black holes
2021
We study the well-motivated mixed dark matter (DM) scenario composed of a dominant thermal WIMP, highlighting the case of $SU(2)_L$ triplet fermion "winos", with a small fraction of primordial black holes (PBHs). After the wino kinetic decoupling, the DM particles are captured by PBHs leading to the presence of PBHs with dark minihalos in the Milky Way today. The strongest constraints for the wino DM come from the production of narrow line gamma rays from wino annihilation in the Galactic Center. We analyse in detail the viability of the mixed wino DM scenario, and determine the constraints on the fraction of DM in PBHs assuming a cored halo profile in the Milky Way. We show that already wi…
Axion star nucleation in dark minihalos around primordial black holes
2020
We consider a general class of axion models, including the QCD and string axion, in which the PQ symmetry is broken before or during inflation. Assuming the axion is the dominant component of the dark matter, we discuss axion star formation in virialized dark minihalos around primordial black holes through gravitational Bose-Einstein condensation. We determine the conditions for minihalos to kinetically produce axion stars before galaxy formation. Today, we expect up to $\sim 10^{17}$ ($\sim 10^9$) axion stars in a radius of 100 parsecs around the Sun for the case of the QCD (string) axion.
JYU-malli : keskitetty julkaisutiedonkeruu Jyväskylän yliopistossa
2020
Observations on the effects of cyclophosphamide, phosphoramide mustard and some activated oxazaphosphorines on murine L1210 leukemia.
1984
The L1210 tumor system was used in vitro and in vivo in comparative studies with activated cyclophosphamide analogs, cyclophosphamide and phosphoramide mustard. All the above compounds gave substantial cell kills (5 logs) of L1210 in vivo at doses that were non-toxic, but slight differences were noted. ASTA Z 7557 had a slight advantage in cure rate over cyclophosphamide when these drugs were given i.v. or i.p. to early tumor (i.p.). However, cyclophosphamide had the advantage in cure rate when drug administration was i.v. to advanced tumor. At equimolar concentrations in vitro ASTA Z 7557 was more cytotoxic than either phosphoramide mustard or acrolein. In vivo, the activated cyclophospham…
Induction of DNA crosslinks and DNA strand lesions by cyclophosphamide after activation by cytochrome P450 2B1
1997
Cyclophosphamide requires metabolic activation by cytochrome P450 to exert its genotoxic effects. Therefore in vitro studies on its mechanism of action have been limited to the use of self-activating derivatives of cyclophosphamide or to hepatocytes as an activating system. In this study we used a cell line of Chinese hamster lung fibroblasts (V79 cells), genetically engineered to express active cytochrome P450 2B1 as the sole observable cytochrome P450 (SD1 cells). An increase in DNA strand lesions (SL: DNA single-strand breaks and alkali labile sites) was observed between 0.5 and 1.5 mM cyclophosphamide (24 h incubation) which could be classified as alkali labile sites using a modified al…
The Translesion Polymerase Rev3L in the Tolerance of Alkylating Anticancer Drugs
2009
Temozolomide and fotemustine, representing methylating and chloroethylating agents, respectively, are used in the treatment of glioma and malignant melanoma. Because chemoresistance of these tumors is a common phenomenon, identification of the underlying mechanisms is needed. Here we show that Rev3L, the catalytic subunit of the translesion DNA polymerase zeta, mediates resistance to both temozolomide and fotemustine. Rev3L knockout cells are hypersensitive to both agents. It is remarkable that cells heterozygous for Rev3L showed an intermediate sensitivity. Rev3L is not involved in the tolerance of the toxic O6-methylguanine lesion. However, a possible role of Rev3L in the tolerance of O6-…
