Asymmetric tandem reactions: New synthetic strategies

The use of domino and multicomponent reactions in asymmetric synthesis is constantly increasing nowadays. This allows for the synthesis of complex molecules in a single synthetic sequence, usually with high atom economy. Herein, we report three examples of new asymmetric tandem reactions recently developed in our laboratories, giving rise to new families of enantiomerically enriched fluorinated and nonfluorinated heterocycles. The reduction of tris-dithiolene complexes of molybdenum(vi) and tungsten(vi) by hydroxide ion: kinetics and mechanism

The kinetic study of the spontaneous reduction of some neutral tris-dithiolene complexes [ML3] of molybdenum(VI) and tungsten(VI), (L = S2C6H42−, S2C6H3CH32− and S2C2(CH3)22−; M = Mo or W) by tetrabutylammonium hydroxide in tetrahydrofuran-water solutions demonstrates that OH− is an effective reductant. Their reduction is fast, clean and quantitative. Depending upon both the molar ratio in which the reagents are mixed and the amount of water present, one- or two-electron reductions of these tris-dithiolene complexes were observed. The Bronsted acid (BA) catalysed Povarov reactions of (E)-1-phenyl-N-(4-(trifluoromethyl)phenyl) methanimine with 1-vinylpyrrolidin-2-one (VPO) and with allyltrimethylsilane (ATMS), affording CF3-substituted cis/trans tetrahydroquinoline (THQ) derivatives, have theoretically been studied using the M06-2X functional together with the standard 6-31G(d) basis set. Abstract The reaction of [1,4-{SiMe3(H)N}2C6Me4] (1) with 2 equivalents of LiBun followed by the addition of SiMe3Cl gave the diamine compound [1,4-{(SiMe3)2N}2C6Me4] (2). [Ta(η5-C5H4SiMe3)Cl4] reacts with 2, in a 2:1 stoichiometric ratio, to initially yield a mixture of the dinuclear, [{Ta(η5-C5H4SiMe3)Cl2}2(μ-1,4-NC6Me4N)] (3), and mononuclear, [Ta(η5-C5H4SiMe3)Cl2{ NC6Me4-4-(N(SiMe3)2)}] (4), imido complexes. 3 can be obtained exclusively by submitting the reaction mixture to repeated cycles of evacuation, to remove volatiles, followed by addition of solvent and subsequent heating. An empirical nucleophilicity index based on the gas-phase ionization potentials has been recently shown to be useful categorizing and settling the nucleophilicity power of a series of captodative ethylenes reacting in cycloaddition reactions (L.R. Explicit Solvation Matters: Performance of QM/MM Solvation Models in Nucleophilic Addition

Nucleophilic addition onto a carbonyl moiety is strongly affected by solvent, and correctly simulating this solvent effect is often beyond the capability of single-scale quantum mechanical (QM) models. This work explores multiscale approaches for the description of the reversible and highly solvent-sensitive nucleophilic N|···C=O bond formation in an Me2N–(CH2)3–CH=O molecule. Synthesis of new asymmetric xanthene dyes via catalyst-free SNAr with sulfur nucleophiles

Addition of a single functional handle to the tricyclic moiety of fluorescein results in asymmetric xanthene dyes. Our synthesis of a new class of asymmetric xanthenes proceeds via an unusual SNAr with sulfur nucleophiles on electron rich aromatic xanthenes scaffolds in the absence of a metal catalyst. The resulting 3'-thioethers exhibit high photostability and are conveniently converted into reactive dyes for macromolecule labelling.
