We developed a non-radioisotopic quantitative competitive RT-PCR method for the measurement of gamma-aminobutyric acid (GABA) type A receptor subunit mRNA levels. The specificity of the method was optimized by the use of four subunit-specific oligonucleotides in the sequential steps: reverse transcription, polymerase chain reaction (PCR), and detection. The biotinylated PCR products were bound on streptavidin-coated microtiter plates allowing detection of the products using dinitrophenyl (DNP)-labeled probes and anti-DNP alkaline phosphatase conjugate. The demixing behavior of 20 representatives of the system oligo(dimethylsiloxane)/oligo(propylene glycol) (ODMS/OPG)1 is investigated at pressures up to 1 500 bar. The degrees of oligomerization range from 2 to 5 for the first and from 1 to 57, 6 for the second component. The experimental results are compared with those for the previously studied system oligoisobutene/oligo(propylene glycol) (OIB/OPG). In both cases the observed upper critical temperatures Tc increase with the number of monomeric units of the less polar component, whereas they run through a minimum when the number of monomeric units of the glycol is raised. A procedure for the separation and identification of small peptides from the water-soluble fraction of a goat cheese was developed. The water-soluble extract was ultrafiltered (1000 Da membrane cutoff), and peptides were isolated by sequential chromatography: size exclusion chromatography (HPLC-grade water), anion exchange chromatography (phosphate buffer gradient), and semipreparative reverse-phase high-performance liquid chromatography (water/acetonitrile gradient). Azobenzene oligomer layers with side chain chromofore groups (ChG) has been experimentally studied to optimize scalar and vector hologram recording at 633 nm. ChG with different acceptors (Br, NO2, H, CN) and different bonding type to the matrix (dispersed without bonding or with covalent bonding) were used. The influence of oligomer matrix (polyvinylbuthirol, tolyle, hexamethylene, polyvinylpirolidone, double matrix) and spacers was also studied. The best results (the maximal diffraction efficiency of 7.9% and the minimal specific recording energy of 0.38 J/(cm2 %)) are obtained for ChG with one NO2 acceptor group covalently bonded to tolyle matrix. Myelin, the multilayered membrane that enwraps and insulates neuronal axons for fast signal propagation, is a plasma membrane specialization of oligodendrocytes and Schwann cells in the central and peripheral nervous system, respectively. Here we provide our lab protocols for the puri fi cation of myelin from mouse brains and for gel-based and gel-free proteomic applications, as well as a brief discussion with respect to our current knowledge of the myelin cytoskeleton.

Dendritic cells (DC) are central regulators of immunity. Signal-induced maturation of DCs is assumed to be the starting point for specific immune responses. To further understand this process, we analyzed the alteration of transcript profiles along the time course of CD40 ligand-induced maturation of human myeloid DCs by Affymetrix GeneChip microarrays covering >6800 genes. Besides rediscovery of genes already described as associated with DC maturation proving reliability of the methods used, we identified clusterin as novel maturation marker. The TAR sequence of the 5' leader of HIV-1 long terminal repeat-directed mRNA was found to be able to bind to and to activate double-stranded RNA-dependent (2'-5')A synthetase. Binding of TAR to the purified synthetase in vitro was abolished by addition of HIV-1 Tat protein, which binds to this sequence with a high affinity. Inhibition of TAR-mediated activation of (2'-5')A synthetase by Tat was prevented in the presence of the Zn2+ and Cd2+ chelators o-phenanthroline and penicillamine, which did not impair TAR-synthetase interaction. Calcium signatures and signaling in cytosol and organelles of tobacco cells induced by plant defense elicitors

Calcium signatures induced by two elicitors of plant defense reactions, namely cryptogein and oligogalacturonides, were monitored at the subcellular level, using apoaequorin-transformed Nicotiana tabacum var Xanthi cells, in which the apoaequorin calcium sensor was targeted either to cytosol, mitochondria or chloroplasts. Our study showed that both elicitors induced specific Ca2+ signatures in each compartment, with the most striking difference relying on duration.
