X-linked Adrenoleukodystrophy (X-ALD) is a rare neurodegenerative disease caused by deficiency of the peroxisomal half-transporter ABCD1, implicated in very long chain fatty acids import. Two additional half-transporters are located in the peroxisomal membrane: ABCD2 and ABCD3. Over-expression of ABCD2 is known to compensate for ABCD1 deficiency, making ABCD2 a therapeutic target for X-ALD treatment. Introduction: Protein Oligomerization and the Formation of Macromolecular Assemblies

The ability of biomolecules to link together to form higher order assemblies underlies much of cellular structure and function. Here we emphasise protein oligomerisation and discuss some of the principles of molecular interaction, from early considerations through to the present day. A few protein examples are presented, selected from our research interests, to highlight assembly features, ranging from the hemoglobins, the hemocyanins to the peroxiredoxins, collagen, the encapsulins and ferritins.

2-Chloro/phenyl-7-arylimino-6,6-dimethylcyclopenta[b]pyridylnickel chlorides: Synthesis, characterization and ethylene oligomerization

Abstract 2-Chloro/phenyl-7-arylimino-6,6-dimethylcyclopenta[b]pyridylnickel chlorides (Ni1–Ni8) were synthesized from the respective ligands L1–L8 and characterized. Upon activation with either methylaluminoxane (MAO) or ethylaluminium sesquichloride (EASC), they show high catalytic activity of up to 10.84 × 106 g(oligomer) mol− 1(Ni) h− 1 in ethylene oligomerization. The products range from butenes to dodecenes for Ni1–Ni4, but are limited to butenes and hexenes in the case of Ni5–Ni8. Ionic Liquids as Homogeneous Catalysts for Glycerol Oligomerization

Ionic liquids (ILs) were used for the first time as catalysts for the glycerin condensation reaction. A series of imidazolium and ammonium ionic liquids differing in the length of the alkyl substituent (C2, C12, and C14) and the type of anion (Br−, CH3COO−, and NaHPO4−) were synthesized using a typical two-step method. The structure of the obtained ILs was confirmed by nuclear magnetic resonance 13C NMR, and their base power was determined on the basis of the Hammett function. Several point mutations have been identified in human aquaporins, but their effects on the function of the respective aquaporins are mostly enigmatic. We analyzed the impact of the aquaporin 2 mutation V71M, which causes nephrogenic diabetes insipidus in humans, on aquaporin structure and activity, using the bacterial aquaglyceroporin GlpF as a model. Importantly, the sequence and structure around the V71M mutation is highly conserved between aquaporin 2 and GlpF. The V71M mutation neither impairs substrate flux nor oligomerization of the aquaglyceroporin. Therefore, the human aquaporin 2 mutant V71M is most likely active, but cellular trafficking is probably impaired.
