Design, Synthesis, and Biological Evaluation of 3,4-Diarylmaleimides as Angiogenesis Inhibitors

The new analogue 2 of combretastatin A-4 was discovered to be an inhibitor of tubulin polymerization with an IC50 of 7.6 microM and reduced angiogenesis in the in vivo chick embryo model. Interestingly, in a series of 2,3-diarylmaleimides closely related to this lead, no other compound was found to be active in the tubulin polymerization assay. However, by screening in the in vivo chick embryo assay 10 was identified as a potent angiogenesis inhibitor indicating an alternative target. Indeed, molecular modeling studies suggest a reasonable binding mode of 10 at the ATP-binding site of the model kinase CDK2. Further advances require new resveratrol analogs be used to identify the structural determinants of resveratrol for the inhibition potency of cell proliferation by comparing experimental and docking studies. Después de haber propuesto que el uso frecuente de singularis en las Verrinas está en buena parte motivado por la alusión aprina que suscitan los nombres parlantes de Verres y de su principal colaborador Apronius, se nos ha opuesto el reparo de que el uso relativo de ese adjetivo es superior en el Pro Sulla. Pues bien, hecho el oportuno análisis, comprobamos que dos de los seis empleos de singularis en este discurso apuntan el mismo valor alusivo. Y los motivos son paralelos: Sulla facilita la conexión con el término genérico del cerdo doméstico o salvaje (sus, suis) y la conducta predatoria del personaje justifi ca la alusión, como en el caso de las Verrinas

A new, simplified photographic method is proposed to estimate the shading effects of obstructions. In spite of the complex mathematical background of the new tool, its practical application is very simple, and only requires the measurements of three angles for each photograph. The proposed method is particularly useful for overcoming the difficulties caused by the need to accurately describe surrounding objects to estimate shading effects. The new method uses the information contained in a picture and allows one to draw daily solar paths directly on the photograph. Synthesis and Biological Evaluation of Combretastatin A-4 and Three Combretastatin-Based Hybrids

The syntheses of combretastatin A-4 from gallic acid and of three combretastatin-based hybrids are described. Starting from commercial gallic acid, the phosphonium salt (3,4,5-trimethoxybenzylphosphonium bromide) is synthesized and coupled, through a Wittig reaction, with several aldehydes, including methoxymethyl-protected isovanillin, the aldehyde γ-bicyclohomofarnesal having a labdane skeleton, 3-(3-pyridyl) propanal, and furfural. The biological properties of the cis-coupled compounds as cytotoxic, antiviral and antifungal agents are also reported. In addition, pyrogallol, gallic and 3,4,5-trimethoxybenzoic acids have been studied biologically.
