Efficient Synthesis of β-Halogeno Protected L-Alanines and Their β-Phosphonium Derivatives.

Abstract Ring opening of oxazolines, prepared from l -serinates, with trimethylsilyl halides (TMSX) led to β-halogeno-N-benzoyl-α-amino esters in good to excellent yields. Quaternization of triphenylphosphine by the β-bromo or -iodo amino esters gave the corresponding β-phosphonium salts in overall yields of up to 93% and with e.e. >96%. Hydrolysis of the ester function afforded the phosphonium salt bearing an N-benzoyl-α-amino acid substituent, with partial racemization. However, the reaction of the TMSX with the carboxylic salt, prepared by saponification of the starting oxazoline ester, furnished the corresponding β-halogeno-N-benzoyl-α-amino acids in 70–95% yields. A rapid and simple procedure was devised for the synthesis of multifunctionalized cyclic β-amino esters and γ-amino alcohols via the 1,3-dipolar cycloaddition of nitrile oxides to β-aminocyclopentenecarboxylates. The opening of the isoxazoline reductive ring to the corresponding highly functionalized 2-aminocyclopentanecarboxylates occurred stereoselectively with good yields.

Five new C2-symmetric chiral ligands of 2,5-bis(imidazolinyl)thiophene (L1–L3) and 2,5-bis(oxazolinyl)thiophene (L4 and L5) were synthesized from thiophene-2,5-dicarboxylic acid (1) with enantiopure amino alcohols (4a–c) in excellent optical purity and chemical yield. The utility of these new chiral ligands for Friedel–Crafts asymmetric alkylation was explored. Subsequently, the optimized tridentate ligand L5 and Cu(OTf)2 catalyst (15 mol%) in toluene for 48 h promoted Friedel–Crafts asymmetric alkylation in moderate to good yields (up to 76%) and with good enantioselectivity (up to 81% ee). A novel phosphorus–carbon bond formation by ring opening with diethyl phosphite of oxazolines derived from serine

Abstract A new reaction of oxazolines derived from serine with diethyl phosphite leading to ring opening products with P–C bond formation is reported. This reaction, which proceeds under neutral conditions and without the use of any halogenated intermediate, results in a mixture of racemic α- and β-phosphono alanines in an approximate 1:2 ratio, with isolated yields up to 77%. The mechanism involves the rearrangement of the oxazoline into the corresponding α-benzamido acrylate, followed by addition of the diethyl phosphite to the double bond. The synthesis of five new oxazoline functionalized vinyl monomers N-[4-(4′,5′-dihydrooxazol-2-yl)phenyl]acrylamide (3 a), N-[4-(4′,5′-dihydrooxazol-2-yl)phenyl]-2-methylacrylamide (3 b), N-{10-[4-(4′,5′-dihydrooxazol-2-yl)phenylcarbamoyl]decyl}-2-acrylamide (5 a), N-{10-[4-(4′,5′-dihydrooxazol-2-yl)phenylcarbamoyl]decyl}-2-methylacrylamide (5 b) and N-[4-(4′,5′-dihydrooxazol-2-yl)-phenyl]-4-vinylbenzamide (7) is described. Specific reactions of amino and carboxyl end groups of Nylon 6 with the reactive oxazoline groups belonging to a cyclophosphazene compound (referred as CP20XA) were carried out at 240 °C for different times, under inert atmosphere. Ny6 polymers terminated with one specific reactive chain end (-COOH or NH2) were reacted with different amounts of CP20XA to study the kinetic order of the reactions. All Ny6-CP20XA reacted products soluble in trifluoroethanol (TFE) were well characterized by MALDI-TOF MS, FT-IR and ( 1H and 13C) NMR techniques. Abstract Oxazoline-amino acids (Xaa-Ozn) occur in natural peptides of potentially important bioactivity. The conformations of the model compounds: Ac-(S)-Ala-Ozn(4R-Me), Ac-(S)-Ala-Ozn(4S–Me), and (gauche+, gauche−, anti) Ac-(S)-Val-Ozn(4R-Me) were studied at meta-hybrid M06-2X/6–311++G(d,p) method including solvent effect. Boc- l -Ala- l -Ozn-4-COOMe and Boc- l -Val- l -Ozn-4-COOMe were synthesized and studied by FT-IR and NMR-NOE methods. The conformations in crystal state were gathered from the Cambridge Structural Data Base. This paper describes the synthesis and characterization of a new class of amphiphilic, water-soluble diblock copolymers based on 2-oxazoline derivatives with pendent N-heterocyclic carbene/palladium catalysts in the hydrophobic block. The synthetic strategy involves a four-step synthesis of three functionalized monomers, each composed of a bis(imidazoline-2-ylidene)palladium(II) diiodide derivative that is covalently linked to a 2-oxazoline monomer via a flexible alkyl spacer (alkyl = butyl, hexyl, octyl). O-Glycoprotein 2-acetamino-2-deoxy-β-d-glucopyranosidase (O-GlcNAcase) hydrolyzes O-linked 2-acetamido-2-deoxy-β-d-glucopyranoside (O-GlcNAc) residues from post-translationally modified serine/threonine residues of nucleocytoplasmic protein. The chemical process involves substrate-assisted catalysis, where two aspartate residues have been identified as the two key catalytic residues of O-GlcNAcase.
