MECHANISMS OF CHROMOSOMAL INSTABILITY: RELATIONSHIP BETWEEN TUMOR SUPPRESSORS AND SAC GENES
2017
p14ARF re-expression induces apoptosis in aneuploid HCT116 cells
2012
Weakening the Spindle Assembly Checkpoint by reduced expression of its components such as MAD2, BubR1 and MPS1 induces chromosome instability and aneuploidy both hallmarks of cancer cells. p14ARF that is found frequently altered in human cancers, is overexpressed in response to oncogenic stimuli to stabilize p53 halting cell progression. Previously, we determined that lack or reduced expression of p14ARF is involved in the maintenance of aneuploid cells suggesting that it could be part of a pathway controlling proliferation of aneuploidy cells. To investigate further this aspect of p14ARF function it was ectopically expressed in HCT116 cells, a stable near diploid cell line, after MAD2 depl…

A Regulatory Mechanism Involving TBP-1/Tat-Binding Protein 1 and Akt/PKB in the Control of Cell Proliferation
2011
Abstract TBP-1 /Tat-Binding Protein 1 (also named Rpt-5, S6a or PSMC3) is a multifunctional protein, originally identified as a regulator of HIV-1-Tat mediated transcription. It is an AAA-ATPase component of the 19S regulative subunit of the proteasome and, as other members of this protein family, fulfils different cellular functions including proteolysis and transcriptional regulation. We and others reported that over expression of TBP-1 diminishes cell proliferation in different cellular contexts with mechanisms yet to be defined. Accordingly, we demonstrated that TBP-1 binds to and stabilizes the p14ARF oncosuppressor increasing its anti-oncogenic functions. However, TBP-1 restrains cell…
Regulation of protein cysteinylation and the inflammatory cascade by thioredoxin-related protein of 14 kDa in pancreas
2020
Although under physiological conditions ROS are essential for normal cell signaling, ROS overproduction under oxidative stress conditions may cause damage to biomolecules. Cysteine residues within proteins are often easily oxidized. Reversible cysteine oxidation is used as a mechanism of redox signaling and control of protein function. However, oxidative stress promotes oxidation of protein thiols, leading to the formation of mixed disulfides between proteins and low-molecular-weight thiols. Disulfide reductases such as thioredoxins can reduce disulfide bonds back to free thiols. Acute pancreatitis is currently one of the leading causes of hospital admission for gastrointestinal disorders, …
Hyvinvointi-, hius- ja kauneusalan ammatteihin opiskelevat kriittisinä nettilukijoina
2023
Tutkimuksessa selvitettiin ammatillisen koulutuksen opiskelijoiden (hyvinvointi-, hius- ja kauneusala) kriittistä nettilukutaitoa. Lisäksi tarkasteltiin aikaisemman tiedon, tekstien lukemisjärjestyksen ja perustelutaitojen yhteyttä siihen, kuinka hyvin opiskelijat osasivat arvioida luotettavia (varmentaminen) ja epäluotettavia (kyseenalaistaminen) nettitekstejä sekä erottaa luotettavat tekstit epäluotettavista. Tutkimukseen osallistui 82 opiskelijaa, joista valtaosa oli 15–18-vuotiaita. Opiskelijat lukivat ja arvioivat kaksi luotettavaa ja kaksi epäluotettavaa tekstiä sokerin vaikutuksista. Opiskelijat arvioivat kirjoittajan asiantuntijuutta, tarkoitusperiä, julkaisupaikkaa ja evidenssin la…
Potent SARS-CoV-2 mRNA Cap Methyltransferase Inhibitors by Bioisosteric Replacement of Methionine in SAM Cosubstrate
2021
Viral mRNA cap methyltransferases (MTases) are emerging targets for the development of broad-spectrum antiviral agents. In this work, we designed potential SARS-CoV-2 MTase Nsp14 and Nsp16 inhibitors by using bioisosteric substitution of the sulfonium and amino acid substructures of the cosubstrate S-adenosylmethionine (SAM), which serves as the methyl donor in the enzymatic reaction. The synthetically accessible target structures were prioritized using molecular docking. Testing of the inhibitory activity of the synthesized compounds showed nanomolar to submicromolar IC50 values for five compounds. To evaluate selectivity, enzymatic inhibition of the human glycine N-methyltransferase invol…
IPR-aineistojen mahdollisuuksia, replikointitarpeita ja teknisiä haasteita
2021
