Desipramine induces disorder in cholesterol-rich membranes:implications for viral trafficking

In this study, the effect of desipramine (DMI) on phospholipid bilayers and parvoviral entry was elucidated. In atomistic molecular dynamics simulations, DMI was found to introduce disorder in cholesterol-rich phospholipid bilayers. This was manifested by a decrease in the deuterium order parameter S(CD) as well as an increase in the membrane area. Disordering of the membrane suggested DMI to destabilize cholesterol-rich membrane domains (rafts) in cellular conditions. The antiphospholipid syndrome (APS) was described in 1983 as a clinical entity characterized by venous and arterial thrombosis, thrombocytopenia, and recurrent fetal loss. The serological markers of APS are antiphospholipid antibodies (APLA) directed mainly against anionic phospholipids, usually cardiolipin but also phosphatidylserine. Some APLA exhibit lupus anticoagulant activity. Monoclonal gammopathy sometimes occurs with the presence of autoantibodies. Antiphospholipid syndrome (APS) is frequently associated with thrombocytopenia, in most cases mild and in the absence of major bleedings. In some patients with a confirmed APS diagnosis, secondary immune thrombocytopenia (ITP) may lead to severe thrombocytopenia with consequent major bleeding. At the same time, the presence of antiphospholipid antibodies (aPL) in patients with a diagnosis of primary ITP has been reported in several studies, although with some specific characteristics especially related to the variety of antigenic targets. Kaninchen uberlebten die letale Dosis von 30 mg/kg Chlorpromazin (i.v.) nur zusammen mit einer Fettinfusion (0,5 ml/kg/min Lipofundin S 10®). Es konnte in vitro gezeigt werden, dass der Zusatz einer Fettemulsion (Lipofundin S 10®) zu Kaninchenblut (25 mg Fett/ml) den Anteil an freiem Chlorpromazin (Gesamtkonzentration 10−4M) von 2,05% auf 0,87% herabsetzt.

Involvement of phospholipid metabolism in the events of the oxidative burst of granulocytes

In summary we provide now evidence for specific interactions between the phospholipid metabolism and the oxidative burst. Lysophosphatidylcholine-like molecules showed specific enhancement of the oxidative burst and the capacity to replace phospholipase A2 function. Our findings might be useful in further studying the events occurring on the way from receptor stimulation to activation of the oxidative burst.

Phospholipase A2, a ubiquitous lipolytic enzyme highly active in the hydrolysis of organized phospholipid substrates, has been characterized optically in its action against a variety of phospholipid monolayers using fluorescence microscopy. By labeling the enzyme with a fluorescent marker and introducing it into the subphase of a Langmuir film balance, the hydrolysis of lipid monolayers in their liquid-solid phase transition region could be directly observed with the assistance of an epifluorescence microscope. Human cytochrome P4501A1 (CYP1A1) is one of the key enzymes in the bioactivation of environmental pollutants such as benzo[a]pyrene (B[a]P) and other polycyclic aromatic hydrocarbons. To evaluate the effect of membrane properties and distinct phospholipids on the activity of human CYP1A1 purified insect cell-expressed human CYP1A1 and of human NADPH-P450 reductase were reconstituted into phospholipid vesicle membranes.
