A Specific Radioimmunoassay for the Determination of Low Quantities of Indole-3-acetic Acid in Spruce Needles of Healthy and Damaged Trees

Summary The aim of the present investigation was to develop a radioimmunoassay for the quantification of indole-3-acetic acid in needles of Picea abies in damaged and phenotypically healthy trees. Phenols, lipophilic substances or compounds with cross-reactivity had to be separated from the extracts in several purification steps. Measurements were carried out between May and October 1986 on trees from two spruce plantations. Marked differences were found in IAA content in needles of healthy and damaged trees. Preparation of extracts from mature spruce needles for enzymatic analyses

It was possible to extract simultaneously several active enzymes involved in the carbohydrate or the amino acid metabolism from spruce needles [Picea abies (L.) Karst.] when a) a 100 mM Na-Pi buffer of pH 7.5 containing 5% PVPP and 0.5% Triton X-100 was used and when b) the resulting crude extracts were freed from lowmolecular-weight compounds by gel-chromatography using the separation medium Fractogel TSK HW-40. Studies on the Content of Free Amino Acids in Needles of Undamaged and Damaged Spruce Trees at a Natural Habitat

Summary Measurements of the amino acids in needles of undamaged and damaged 90-year-old spruce trees ( Picea abies ) showed different patterns. During the summer months a reduction in total amino acid content of about 60 % was found. In autumn the content of total amino acids rose again. The courses of the single amino acids behave similarly to that of the total amino acids. Quantitatively, the main amino acids were GLU, ASP and GLN with about 37 %, 21 % and 10 %, respectively, followed by PRO, ALA and SER with about 3–4 % each. Summary The diurnal courses of the contents of 1-aminocyclopropane carboxylic acid (ACC), its malonyl conjugate (MACC) and abscisic acid (ABA) in the two-year-old needles of damaged and undamaged spruce trees (Picea abies L.) were investigated. The contents of ACC and MACC were significantly higher in the needles of the damaged trees as compared with the undamaged ones. In the needles of the damaged spruce trees, the ACC and MACC contents, and ABA contents as well, fluctuated to a greater extent than those in the undamaged spruce trees during the day. The relationships between ACC-ABA and MACC-ACC contents as well are also discussed.

Diagnosis of Damage to Norway Spruce (Picea Abies) through Ethylene and Other Biochemical Criteria

Forest damage normally is characterized by loss and yellowing of leaves or needles. The results of this ocular damage diagnosis can be confirmed and stated more precisely through biochemical criteria, e.g. the plant hormone ethylene, the activity of the enzyme phosphoenolpyruvate-carboxylase (PEP-C), the content of chlorophyll and other biochemical parameters. Therefore we investigated the interrelation between visible damage and 13 biochemical parameters in six trees each from eleven permanent observed sites in the south-west of Germany. Abstract The content of several phenolic compounds in needles of 20- to 30-year-old Norway spruce trees (Picea abies) was measured using HPLC. The results of two forestry sites in middle western Germany are reported in this paper. They are part of a research programme on novel forest decline which was carried out in various regions of Germany. Distinct amounts of picein, catechin, piceatannol glucoside, and other phenolic compounds were detected in the studied spruce needles. Additionally, their contents changed in relation to damage. Many protected areas have been under intensive forest management prior to protection and thus lack natural ecosystem structures and dynamics. Dead wood is a key structure in forests harboring hundreds of threatened species. We investigated the ecological success of dead wood creation as a boreal forest restoration measure. Macro Element Content in Fertilized Forest Soils in Spruce (Picea Abies (L.) H. Karst.) Stands in Latvia

The present paper provides an overview of analysis conducted with soil samples from depths up to 80 cm, obtained in the Klive area from a spruce stand owned by JSC “Latvijas Valsts meži”. Also test fertilization attempts were made with the use of ash- and potassium-based artificial fertilizers (K2SO4). The content of macro elements (K, Ca, N, and C) was determined in the soil samples in the course of the analysis. The obtained results indicate that the content of K and Ca decreases at greater horizontal depths of the soil. Nutrients are subsequently being leached away from the soil.
