A new actor involved in hypothalamic glucose detection : the Transient Receptor Potential Canonical (TRPC) channels

Hyperglycemia is detected and integrated by the mediobasal hypothalamus (MBH) which, in turn, inhibits food intake and triggers insulin secretion. The MBH houses specialized glucose-sensitive (GS) neurons, which directly or indirectly modulate their electrical activity in response to changes in glucose level. In a first study, we hypothesized that indirect detection of glucose by MBH GS neurons involves the secretion of endozepine by astrocytes, a gliotransmitter known to inhibit food intake in response to hyperglycemia. Lipolysis in isolated epididymal adipocytes from genetically obese Zucker rat treated with 3,5,3'-L-triiodothyronine.

Abstract. Glycerol released by isolated epididymal adipocytes was measured in 6-8 weeks old genetically obese (fa/fa) and lean (C.c.a/-) male Zucker rats, ip treated or not treated with T3 (0.46, 2.3 and 4.6 nmol/100 g in a single injection or 4.6 nmol · (100 g)−1 · day−1 for 5 days). In the non-treated rats, lipolysis was less stimulated by adrenalin or isoproterenol in obese than in lean rats, whereas it was stimulated by theophylline up to the same level in both kinds of rats. The oldest rocks of Greece: first evidence for a Precambrian terrane within the Pelagonian Zone

The Pelagonian Zone in Greece represents the westernmost belt of the Hellenide hinterland (Internal Hellenides). Previous geochronological studies of basement rocks from the Pelagonian Zone have systematically yielded Permo-Carboniferous ages. In this study we demonstrate, for the first time, the existence of a Precambrian crustal unit within the crystalline basement of the Pelagonian Zone. The U–Pb single-zircon and SHRIMP ages of these orthogneisses vary from 699 ± 7 Ma to 713 ± 18 Ma, which identify them as the oldest rocks in Greece. Pineal 'synaptic' ribbon numbers and melatonin synthesis of rat are resistant to guanethidine sympathectomy.

Chemical sympathectomy is widely used to study the impact of the noradrenergic system on neuronal and neuroendocrine circuits. We tested the effects of intraperitoneal injections of guanethidine, an adrenergic neuron blocking agent, on selected functional parameters of the rat pineal gland which are known to be under sympathetic influence. The reliability of the method was demonstrated by the clear enophthalmus developed by experimental animals. However, neither the numbers of 'synaptic' ribbons nor melatonin synthesis differed between treated and control rats, both parameters exhibiting the nocturnal increase seen in intact animals. Co-transmitter mediated facilitation by sympathetic nerve stimulation of evoked acetylcholine release from the rabbit perfused atria preparation.

Rabbit atria were isolated with the extrinsic right sympathetic and vagus nerves attached and perfused with Tyrode solution. Acetylcholine overflow was determined after labelling of the transmitter stores with [14C]choline and fractionation of the radioactivity on cation exchange columns. Sympathetic nerve stimulation (SNS, 2 Hz, 3 min) carried out together with vagus nerve stimulation (VNS, 2 Hz, 3 min), but each SNS pulse preceding a vagal one by 19 ms, caused a facilitation of acetylcholine overflow of about 60% versus independent controls in the absence of SNS. Synthesis and induction of G0–G1 phase arrest with apoptosis of 3,5-dimethyl-6-phenyl-8-(trifluoromethyl)-5,6-dihydropyrazolo[3,4-f][1,2,3,5]tetrazep…

The multistep synthesis of 3,5-dimethyl-6-phenyl-8-(trifluoromethyl)-5,6-dihydropyrazolo[3,4-f][1,2,3,5]tetrazepin-4(3H)-one 15 has been carried out. The compound showed antiproliferative and apoptotic effects against K562, K562-R (imatinib mesilate resistant), HL60 and multidrug resistant (MDR) HL60 cell lines. Compound 15 showed a pro-apoptotic activity against HL60 and K562 resistant cell lines markedly higher than etoposide and busulfan, respectively. Flow cytometry studies carried out on K562 cells allowed to establish that 15 induces G0-G1 phase arrest followed by apoptosis.

Objectives: We aimed to assess the effect of lumbar spine fusion (LSF) on disability, health-related quality of life and mortality in a 5-year follow-up, and to compare these results with the general population. Methods: 523 consecutive LSF operations were included in a prospective follow-up.
