A Library-Based Screening Strategy for the Identification of DARPins as Ligands for Receptor-Targeted AAV and Lentiviral Vectors

Delivering genes selectively to the therapeutically relevant cell type is among the prime goals of vector development. Here, we present a high-throughput selection and screening process that identifies designed ankyrin repeat proteins (DARPins) optimally suited for receptor-targeted gene delivery using adeno-associated viral (AAV) and lentiviral (LV) vectors. In particular, the process includes expression, purification, and in situ biotinylation of the extracellular domains of target receptors as Fc fusion proteins in mammalian cells and the selection of high-affinity binders by ribosome display from DARPin libraries each covering more than 1012 variants. Symptomatic accommodative and binocular dysfunctions from the use of flat-panel displays

AIM: To determine the presence of symptomatic accommodative and non-strabismic binocular dysfunctions (ANSBD) in a non-presbyopic population of video display unit (VDU) users with flat-panel displays. METHODS: One hundred and one VDU users, aged between 20 to 34y, initially participated in the study. This study excluded contact-lens wearers and subjects who had undergone refractive surgery or had any systemic or ocular disease. Getting into the Same Boat – Enabling the Realization of the Disabled Child’s Agency in Adult–Child Play Interaction

The purpose of this study was to find out how an adult can enable or hinder the realization of a disabled child’s agency in play interaction. We focused on the child’s play invitations, which were constructed as dispreferred by the adult. The data consisted of nine videotaped playing situations with five nurses and five disabled children in a children’s neurological ward. The microanalysis with interventionist applied conversation analysis focused on one playing situation between one nurse and one three-year-old boy with no spoken language. The effects of playing tactics on creating scoring opportunities in random matches from US Major League Soccer

The aim of this study was to examine the effects of playing tactics on the creation of scoring opportunities in random matches from US Major League Soccer 2014, taking into account both contextual and defensive variables. The sample included 3253 team possessions corresponding to 30 randomly selected matches. Multidimensional qualitative data from 10 categorical variables was analysed by means of descriptive analysis and multiple logistic regression analysis. Contextual variables such as match location (p=0.049), match half (p=0.043) and match status (P=0.032) were associated with creating scoring opportunities. Body coloration and mechanisms of colour production in Archelosauria: the case of deirocheline turtles

Animal body coloration is a complex trait resulting from the interplay of multiple mechanisms. While many studies address the functions of animal coloration, the mechanisms of colour production still remain unknown in most taxa. Here we compare reflectance spectra, cellular, ultra- and nano-structure of colour-producing elements, and pigment types in two freshwater turtles with contrasting courtship behaviour,Trachemys scriptaandPseudemys concinna. Enhanced field-of-view integral imaging display using multi-Köhler illumination.

A common drawback in 3D integral imaging displays is the appearance of pseudoimages beyond the viewing angle. These pseudoimages appear when the light rays coming from each elemental image are not passing through the corresponding microlens, and a set of barriers must be used to avoid this flipping effect. We present a pure optical arrangement based on Köhler illumination to generate these barriers thus avoiding the pseudoimages. The proposed system does not use additional lenses to project the elemental images, so no optical aberrations are introduced. Despite of the improved therapeutic strategies, the survival rate of high-risk neuroblastoma patients is poor.
