Improved stability of black lipid membranes by coating with polysaccharide derivatives bearing hydrophobic anchor groups

1986

Abstract Black lipid membranes were coated with modified polysaccharides bearing hydrophobic palmitoyl and cholesteryl moieties. The changes in membrane structure were investigated using dipicrylamine, a lipophilic ion, as membrane probe. The kinetics of ion transport through the black lipid membranes were studied using the charge pulse relaxation technique. With this technique it was found that it is possible to detect the insertion of the hydrophobic anchor groups of the polysaccharides into the membrane bilayer. As a result of the surface coating, these membranes exhibit a drastically increased long-term stability.

Enhanced Activity and Stereoselectivity of Polystyrene-Supported Proline-Based Organic Catalysts for Direct Asymmetric Aldol Reaction in Water

2009

Several polystyrene-supported proline dipeptides and a prolinamide derivative were prepared by thiol–ene coupling. These materials were used as catalysts for the direct asymmetric aldol reaction in water, and results compared with unsupported catalysts in water. Such an approach gave more active or stereoselective catalysts compared to the unsupported compounds, showing that our immobilization procedure may be useful to develop catalytic materials with enhanced performance. Moreover, these catalysts can be recovered and reused for at least nine times without loss of activity or can be easily regenerated when their activity has decreased. (© Wiley-VCH Verlag GmbH & Co. ChemInform Abstract: Synthesis and in vitro Studies on a Potential Dopamine Prodrug.

2009

Dopamine delivery to the central nervous system (CNS) undergoes the permeability limitations of blood-brain barrier (BBB) which is a selective interface that excludes most water-soluble molecules from entering the brain. Neutral amino acids permeate the BBB by specific transport systems. Condensation of dopamine with neutral amino acids could afford potential prodrugs able to interact with the BBB endogenous transporters and easily enter the brain. The synthesis and characterization of the dopamine derivative 2-amino-N-[2-(3,4-dihydroxy-phenyl)-ethyl]-3-phenyl-propionamide (7) is described. The chemical and enzymatic stability of 7 was evaluated. The molecular weight (300 Da) and Log Papp (…

Convective electrodiffusion processes through graft-modified charged porous membranes

1999

Convective diffusion and electrophoresis across a charged porous membrane showing variable permeability properties were studied. The membrane used was prepared by grafting poly(acrylic acid) (PAA) onto a porous polyvinylidene fluoride (PVDF) membrane. The degree of grafting was selected to be relatively low, 18 wt%, to compromise between the ion selectivity and hydraulic permeability of the membrane. The possible electric field induced effects on the membrane permselectivity were examined after the membrane had been characterized by convective diffusion and concentration cell potential measurements. The measured electrophoresis data, however, could be modelled using the extended Teorell–Mey…

Kinetic modeling of ion conduction in KcsA potassium channel.

2005

KcsA constitutes a potassium channel of known structure that shows both high conduction rates and selectivity among monovalent cations. A kinetic model for ion conduction through this channel that assumes rapid ion transport within the filter has recently been presented by Nelson. In a recent, brief communication, we used the model to provide preliminary explanations to the experimental current-voltage J-V and conductance-concentration g-S curves obtained for a series of monovalent ions (K(+),Tl(+), and Rb(+)). We did not assume rapid ion transport in the calculations, since ion transport within the selectivity filter could be rate limiting for ions other than native K(+). This previous wor…

Recyclable Catalyst Reservoir: Oxidation of Alcohols Mediated by Noncovalently Supported Bis(imidazolium)-Tagged 2,2,6,6-Tetramethylpiperidine 1-Oxyl

2013

Bis(imidazolium)-tagged 2,2,6,6-tetramethylpiperidine 1-oxyl (TEMPO) catalysts were adsorbed on different supports such as silica gel, silica gel modified with highly cross-linked polymeric imidazolium networks, and highly cross-linked polymeric imidazolium networks entrapping magnetic particles. These systems provided a convenient tool for the oxidation of both primary and secondary alcohols working as recyclable reservoirs for the bis(imidazolium)-tagged TEMPO catalysts. By using EPR spectroscopy it was demonstrated that the catalyst was released as the corresponding oxoammonium salt in the solution during the recycling step, thus promoting the oxidative process in a homogeneous fashion. …

Dual effects of increased glycogen synthase kinase-3β activity on adult neurogenesis

2013

Adult neurogenesis, the generation of new neurons during the adulthood, is a process controlled by several kinases and phosphatases among which GSK3β exerts important functions. This protein is particularly abundant in the central nervous system, and its activity deregulation is believed to play a key role in chronic disorders such as Alzheimer's disease. Previously, we reported that in vivo overexpression of GSK3β (Tet/GSK3β mice) causes alterations in adult neurogenesis, leading to a depletion of the neurogenic niches. Here, we have further characterized those alterations, finding a delay in the switching-off of doublecortin marker as well as changes in the survival and death rates of imm…

Calcium signatures and signaling in cytosol and organelles of tobacco cells induced by plant defense elicitors

2011

Calcium signatures induced by two elicitors of plant defense reactions, namely cryptogein and oligogalacturonides, were monitored at the subcellular level, using apoaequorin-transformed Nicotiana tabacum var Xanthi cells, in which the apoaequorin calcium sensor was targeted either to cytosol, mitochondria or chloroplasts. Our study showed that both elicitors induced specific Ca2+ signatures in each compartment, with the most striking difference relying on duration. Common properties also emerged from the analysis of Ca2+ signatures: both elicitors induced a biphasic cytosolic [Ca2+] elevation together with a single mitochondrial [Ca2+] elevation concomitant with the first cytosolic [Ca2+] p…
