Proposal of a Citrus translational genomic approach for early and infield detection of Flavescence dorée in Vitis
2014
Flavescence dore´e (FD) is one of the most widely known grapevine yellows disease and one of the most unabated worldwide in the viticulture sector. In this paper, we outline a strategy for developing an integrated system of technologies to enable rapid, early disease FD detection and diagnosis. We propose the deployment of a newly developed sensor device, the differential mobility spectrometer (DMS), which has shown positive results with a similar vector-borne disease in Citrus. We have previously demonstrated that the gas chromatograph DMS (GC/DMS) can distinguish various citrus diseases, and the system may also allow detection of volatile organic compound (VOC) signals from a tree of othe…
In situ Phenotyping of Grapevine Root System Architecture by 2D or 3D Imaging: Advantages and Limits of Three Cultivation Methods
2021
International audience; The root system plays an essential role in the development and physiology of the plant, as well as in its response to various stresses. However, it is often insufficiently studied, mainly because it is difficult to visualize. For grapevine, a plant of major economic interest, there is a growing need to study the root system, in particular to assess its resistance to biotic and abiotic stresses, understand the decline that may affect it, and identify new ecofriendly production systems. In this context, we have evaluated and compared three distinct growing methods (hydroponics, plane, and cylindric rhizotrons) in order to describe relevant architectural root traits of …
Autotetraploid Emergence via Somatic Embryogenesis in Vitis vinifera Induces Marked Morphological Changes in Shoots, Mature Leaves, and Stomata
2021
Polyploidy plays an important role in plant adaptation to biotic and abiotic stresses. Alterations of the ploidy in grapevine plants regenerated via somatic embryogenesis (SE) may provide a source of genetic variability useful for the improvement of agronomic characteristics of crops. In the grapevine, the SE induction process may cause ploidy changes without alterations in DNA profile. In the present research, tetraploid plants were observed for 9.3% of ‘Frappato’ grapevine somatic embryos regenerated in medium supplemented with the growth regulators β-naphthoxyacetic acid (10 µM) and N6-benzylaminopurine (4.4 µM). Autotetraploid plants regenerated via SE without detectable changes in the …
Selection of indigenous yeast strains for the production of sparkling wines from native Apulian grape varieties.
2018
We report the first polyphasic characterization of native Saccharomyces cerevisiae in order to select candidate strains for the design of starter cultures tailored for Apulian sparkling wines obtained from local grape variety. In addition, it is the first survey in our region that propose the selection of autochthonous starter cultures for sparkling wine i) including a preliminary tailored genotypic and technological screening, and ii) monitoring analytical contribution during secondary fermentation in terms of volatile compounds (VOCs). Furthermore, we exploit the potential contribute of autochthonous cultures throughout the productive chain, including the possible improvement of base wine…
Survey of five major grapevine viruses infecting Blatina and Žilavka cultivars in Bosnia and Herzegovina
2021
The sanitary status of grapevines has not yet been considered sufficiently in vineyards throughout Bosnia and Herzegovina (BiH). An extensive survey of five major grapevine viruses in the country was carried out in 2019. A total of 630 samples from the two dominant autochthonous cultivars, named Žilavka and Blatina, were tested by DAS-ELISA for the presence of grapevine leafroll-associated viruses (GLRaV-1 and 3), grapevine fleck virus (GFkV), grapevine fanleaf virus (GFLV) and Arabis mosaic virus (ArMV). Eighty-eight % of the samples were positive for at least one virus, and all five viruses were detected, thought with different incidence, i.e. GLRaV-3 (84%), GFLV (43%), GLRaV-1 (14%), GFk…
Berries variability: causes and effects on the quality of ‘Cabernet Sauvignon’
2018
In this study the objective was to investigate if differences in berry size (within a population of berries from different bunches) and flowering process would lead to differences in qualitative parameters and berries variability. The trial was conducted during the 2009 vegetative season at an irrigated ‘Cabernet Sauvignon’/1103 P vineyard. At the beginning of flowering, for 100 inflorescences, all open flowers were marked by red ink while 5 days later, closed flowers were marked by black ink. Open and closed flower distribution was investigated along the rachis. At pea size, 20 marked bunches were sampled. The diameter and weight of all berries was measured and Gaussian distribution was pe…
Clone-Dependent Expression of Esca Disease Revealed by Leaf Metabolite Analysis
2019
International audience; Grapevine trutk diseases, especially Esca, are of major concern since they gradually alter vineyards worldwide and cause heavy economic losses. The expression of Esca disease symptoms depends on several factors, including the grapevine cultivar. In this context, a possible clone-dependent expression of the Esca disease was studied. Two clones of 'Chardonnay' grown in the same plot were compared according to their developmental and physiological traits, metabolome, and foliar symptom expression. Analysis of their leaf metabolome highlighted differences related to symptom expression. Interestingly, the content of a few specific metabolites exhibited opposite variations…
An ancestral allele of grapevine transcription factor MYB14 promotes plant defence
2016
Highlight The molecular mechanisms underlying the elevated inducibility of stilbene in pathogen-resistant Vitis sylvestris can be explained by the increased inducibility of the MYB14 promoter.
The Influence of Film and Storage on the Phenolic and Antioxidant Properties of Red Raspberries (Rubus idaeus L.) cv. Erika
2019
In this paper, the effect of the packaging material and storage method on red raspberries produced at Reggio Calabria (Italy) was studied. For this purpose, the fruits were stored immediately after harvest in different conditions: in the fridge at 1 °
Combined influence of bud load and bud position along the cane on vegetative and reproductive parameters of grape cv. Grillo
2019
Cultivar Grillo vines are characterized by problems with flower biology (the fertility of basal buds) and fructification (millerandage). In this study, to manage the variability in bunch weight with winter pruning and to program others canopy management practices (i.e. early defoliation), three different treatments of bud load were set up by leaving the cane with 3, 6 or 10 buds. The effects of bud load and cane length were studies regarding bud fertility, shoot leaf area, and the number of flowers and berries, as well as the relationship between leaf area and percentage of fruit set, leaf area/flower and percentage of fruit set, and the number of hens and chicks berries. Shoots in the dist…
