A FRET-based random screening assay was used to generate hit compounds as sortase A inhibitors that allowed us to identify ethyl 3-oxo-2-(2-phenylhydrazinylidene)butanoate as an example of a new class of sortase A inhibitors. Other analogues were generated by changing the ethoxycarbonyl function for a carboxy, cyano or amide group, or introducing substituents in the phenyl ring of the ester and acid derivatives. The most active derivative found was 3-oxo-2-(2-(3,4dichlorophenyl)hydrazinylidene)butanoic acid (2b), showing an IC50 value of 50 µM. TP53 mutations and S-phase fraction but not DNA-ploidy are independent prognostic indicators in laryngeal squamous cell carcinoma

To prospectively evaluate the prognostic significance of TP53, H-, K-, and N-Ras mutations, DNA-ploidy and S-phase fraction (SPF) in patients affected by locally advanced laryngeal squamous cell carcinoma (LSCC). Eight-one patients (median follow-up was 71 months) who underwent resective surgery for primary operable locally advanced LSCC were analyzed. Tumor DNA was screened for mutational analysis by PCR/SSCP and sequencing. DNA-ploidy and SPF were performed by flow cytometric analyses. Sponges (Porifera) represent the lowest metazoan phylum, characterized by a pronounced plasticity in the determination of cell lineages, and they are the closest related taxon to the hypothetical ancestral animal, the Urmetazoa, from which the metazoan lineages diverged. In a first approach to elucidate the molecular mechanisms controlling the switch from the cell lineage with a putative indefinite growth capacity to senescent, somatic cells, the activity of the telomerase as an indicator for immortality has been determined. Molecular analysis of lichen-associated bacterial communities.

The bacterial communities associated with 11 different lichen samples (belonging to eight different species) from different habitats were investigated. The culturable aerobic-heterotrophic fraction of the bacterial communities was isolated from nine lichen samples on protein-rich and sugar-rich/N-free media. Thirty-four bacterial isolates were purified and pooled into groups (phylotypes) by analysis of the ribosomal internal transcribed spacer polymorphism. The article discusses selected lexical units found in the dialects of Silesia, which refer to the act of speaking. It examines more than 190 tokens and collocations whose common denominator is the meaning of ‘speaking’. Analyzed are units referring to the content of the expression (with the main semantic component ‘to talk nonsense’), as well as those referring to physical aspects of the act of speaking, such as the clarity and accuracy of articulation (‘to speak indistinctly’, ‘to stutter’), the pace of speaking (‘to speak fast’, ‘to speak slowly’), also the tone, pitch, and intensity of the voice (‘to speak softly’, ‘to say shrilly’). The aim of the article is to show how to list and develop phraseology in the oldest Polish dialect dictionary: A Dictionary of Polish Dialects by Jan Karłowicz, and then discuss the placement of phraseology in the macro- and microstructure of later, 20th century selected dialectal dictionaries. Therefore, the article is an overview of the tradition of lexical development and study of phraseology in dialectal dictionaries (on the example of Silesian Dialect Dictionary, Polish Dialects Dictionary by Polish Academy of Sciences, the dictionary of Bernard Sychta, Dictionary of Ostróda, Warmia and Mazury Dialects).

The aim of this article is to research the ways of developing phraseology and its emplacement into the macro- and microstructure in selected dialectal dictionaries (academic and popular). The first part is an overview of the phraseology, as included in introductions to the dictionaries. In the second part, using specific examples, the author discusses how the methodological and editorial assumptions adopted by dictionary authors/editors are followed in practice. Functional characterization of the human tRNA methyltransferases TRMT10A and TRMT10B

Abstract The TRM10 family of methyltransferases is responsible for the N1-methylation of purines at position 9 of tRNAs in Archaea and Eukarya. The human genome encodes three TRM10-type enzymes, of which only the mitochondrial TRMT10C was previously characterized in detail, whereas the functional significance of the two presumably nuclear enzymes TRMT10A and TRMT10B remained unexplained. Here we show that TRMT10A is m1G9-specific and methylates a subset of nuclear-encoded tRNAs, whilst TRMT10B is the first m1A9-specific tRNA methyltransferase found in eukaryotes and is responsible for the modification of a single nuclear-encoded tRNA. International audience; Modified nucleotide 5-methylcytosine (m(5)C) is frequently present in various eukaryotic RNAs, including tRNAs, rRNAs and in other non-coding RNAs, as well as in mRNAs. RNA: m(5)C-methyltranferases (MTases) Nop2 from S. cerevisiae and human proliferation-associated nucleolar antigen p120 are both members of a protein family called Nop2/NSUN/NOL1. Protein p120 is well-known as a tumor marker which is over-expressed in various cancer tissues. Użytkownicy internetu korzystają z możliwości, jakie daje im komputer, i chętnie tworzą teksty łączące w sobie elementy werbalne i graficzne. Jednym z najpopularniejszych polikodowych gatunków internetowych są demotywatory. Połączenie w nich dwóch kanałów: werbalnego i ikonicznego powoduje wzmocnienie siły ich oddziaływania. Często w werbalnej części demotywatorów są wykorzystywane frazeologizmy i skrzydlate słowa. Połączenie ich z elementem graficznym powoduje ich defrazeologizację oraz grę językową z wykorzystaniem wieloznaczności i homonimiczności wyrazów.
