Consensus modelling and molecular dynamics studies for the identification of novel telomerase inhibitors as anti-cancer agents

Telomerase plays an important role in early stages of life-maintaing telomere and chromosomal integrity of frequently dividing cells. It turns dormant in most somatic cells during adulthood. However, in cancer cells, telomerase gets reactivated and works tirelessly to maintain the length of telomeres, leading to immortality of cells. Hence, in this study, we have used a combined ligand-based and structure based drug design approach for the identification of novel telomerase inhibitors as anti-cancer agents. Synthesis, antiproliferative activity, and in silico insights of new 3-benzoylamino-benzo[ b ]thiophene derivatives

A new series of 3-benzoylamino-5-imidazol-5-yl-benzo[b]thiophenes and the parent amino derivatives were synthesized and screened as antitumor agents. All tested compounds showed concentration-dependent antiproliferative activity profile against HeLa cell line, exhibiting GI50 values in the low micromolar range. The most active compounds were tested in cell cycle perturbation experiments. A rapid accumulation of cells in the G2/M phase, with a concomitant reduction of cells in both the S and G0/G1 phases, was observed, suggesting that cell exposure to selected derivatives produces mitotic failure. Kazoo training for L2 pronunciation practice and reduced foreign accentedness?

This paper presents a controlled pretest–training–posttest study on using a small membraphone instrument called kazoo for L2 pronunciation practice and reducing foreign accentedness. Learners were recorded for free speech before and after a six-week pronunciation and oral skills course including L2 prosody training with personal kazoos. A control group did not receive any kazoo training but did similar tasks in regular speech. Samples of all learners’ free speech before and after the course were rated for foreign accentedness by L1 speakers. New is not always costly: evidence from online processing of topic and contrast in Japanese.

Two visual ERP experiments were conducted to investigate topic and contrast assigned by various cues such as discourse context, sentential position and marker during referential processing in Japanese. Experiment 1 showed that there was no N400-difference for new vs. given noun phrases (NPs) when the new NP was expected (contrastively focused) based on its preceding context and sentential position. Experiment 2 further revealed that the N400 for new NPs can be modulated by the NP’s contrastive meaning (exhausitivity) induced from the marker. Human Topoisomerase I interaction with DNA on flexible nylon substrate for malaria diagnosis

The study of interaction between human Topoisomerase I (hTopo I) and the appropriate sequences of DNA is shown, with the aim to realize a biosensor for malaria diagnosis. Topoisomerases alter the topological state of DNA by catalysing the breaking and rejoining of DNA strands1. The ligase activity is fundamental for our biosensing strategy, revealing the pathology. hTopo I was chosen as a model system for the Plasmodium analogous. The biosensor is a new flexible and easy to degrade biochip printed on nylon by dip-pen lithography (DPL). Double copies of blaKPC-3::Tn4401a on an IncX3 plasmid in Klebsiella pneumoniae successful clone ST512 from Italy

ABSTRACT A carbapenem-resistant sequence type 512 (ST512) Klebsiella pneumoniae carbapenemase 3 (KPC-3)-producing K. pneumoniae strain showing a novel variant plasmid content was isolated in Palermo, Italy, in 2014. ST512 is a worldwide successful clone associated with the spread of bla KPC genes located on the IncFIIk pKpQIL plasmid. In our ST512 strain, the bla KPC-3 gene was unusually located on an IncX3 plasmid, whose complete sequence was determined. Two copies of bla KPC-3 ::Tn 4401a caused by intramolecular transposition events were detected in the plasmid.

Summary: Chromatin organization plays a crucial role in tissue homeostasis. Heterochromatin relaxation and consequent unscheduled mobilization of transposable elements (TEs) are emerging as key contributors of aging and aging-related pathologies, including Alzheimer’s disease (AD) and cancer. However, the mechanisms governing heterochromatin maintenance or its relaxation in pathological conditions remain poorly understood.
