URBAN REGENERATION AND BUILT HERITAGE

2021

The rehabilitation project is mainly addressed to the adaptation of new housing and urban needs, but it is obviously conditioned and/or “constrained” by the limitation of the building itself, in particular when its intrinsic characteristics must be preserved. In this paper we focus on some aspects of the general framework of urban regeneration: the balance on the “urban question” and on the methodology of integrated planning; the connection between quality and urban liveability; the sharing of the objectives of regeneration and its meanings. Without claiming to be exhaustive, some of the “enabling” conditions and the “indispensable” reasons and conditions for guiding the process of urban re…

Modelling the regenerative niche: a major challenge in biomaterials research

2015

By definition, biomaterials are developed for clinical application. In the field of regenerative medicine their principal function is to play a significant, and, if possible, an instructive role in tissue healing. In the last analysis the latter involves targeting the 'regenerative niche'. The present paper will address the problem of simulating this niche in the laboratory and adopts a life science approach involving the harnessing of heterotypic cellular communication to achieve this, that is, the ability of cells of different types to mutually influence cellular functions. Thus, co-culture systems using human cells are the methodological focus and will concern four exemplary fields of re…

In vitro Shoot Regeneration from Flower and Leaf Explants in Rhododendron

2003

Rhododendron shoot regeneration was accomplished using either flower explants (each consisting of ovary with pedicel) of Rhododendron cvs. Nova Zembla and Irina or leaves isolated from in vitro grown Rhododendron catawbiense Michx. Multiple shoot tip clumps were obtained on Anderson's medium containing 0.5 to 1.5 mg dm−3 thidiazuron (TDZ) in combination with 12 to 15 mg dm−3 N6-[2-isopentenyl]adenine (2iP) and 1 to 3 mg dm−3 indole-3-butyric acid (IBA). After 16 weeks on the regeneration media, explants with shoot tip clumps were transferred for shoot elongation to Anderson's medium with 3 mg dm−3 2iP. Two months later, the shoots have reached 5 to 40 mm in length and were fit for subcultiv…

From limit to centrality. Regeneration of the Genoa's port area through architectural and urban design

2012

This survey’s core is the case study of the regeneration of the port area and of the urban waterfront of the city of Genoa, as an emblematic example where the architectural and urban design plays an important role, oriented to the transformation of the port area and to the modification of the relationship that exists between the harbour and the city. In particular this will deepen the possibility, to reverse the condition of limit that characterizes the port area into a new centrality, through the design, passing by intermediate steps that redefine the modalities of access and of connection between city and port, breaking the condition of isolation that ports usually present. As a matter of…

Resilient cities and sustainable urban mobility in Sicily

2018

The global crisis that in recent years has been investing our planet at different levels - social, economic, political, cultural, environmental - has determined a constant change in the quality of life in the cities. Cities, unfortunately, increasingly vulnerable characterized by insecurity, uncertainty, unpredictability due to mistakes made in the past. This is why, today, urban planning is called to satisfy the new needs of the community through a planning that must be adapted to change. In this scenario, urban centers - in order to ensure high levels of quality of life and responsible management of territorial resources - they must re-examine their governance, their strategies for expans…

Inducible and reversible inhibition of miRNA-mediated gene repression in vivo

2021

Although virtually all gene networks are predicted to be controlled by miRNAs, the contribution of this important layer of gene regulation to tissue homeostasis in adult animals remains unclear. Gain and loss of function experiments have provided key insights into the specific function of individual miRNAs, but effective genetic tools to study the functional consequences of global inhibition of miRNA activity in vivo are lacking. Here we report the generation and characterization of a genetically engineered mouse strain in which miRNA-mediated gene repression can be reversibly inhibited without affecting miRNA biogenesis or abundance. We demonstrate the usefulness of this strategy by invest…

Genomic response programs of Candida albicans following protoplasting and regeneration

2005

Transcription profiling of Candida albicans cells responding to the elimination of the wall (protoplasts) and posterior regeneration was explored. DNA microarrays were used to measure changes in the expression of 6039 genes, and the upregulated genes during regeneration at 28 degrees C were assigned to fourteen categories. A total of 407 genes were upregulated during the process, of which 144 reached a maximum after 1 h. MKC1, a gene encoding a member of the regulatory pathway involved in cell wall integrity was overexpressed. Time-dependent expression divided the genes into 40 clusters. Clusters 1-19 were highly expressed initially (time 0) and downregulated following incubation, whereas t…

O-102 Polymeric scaffold loaded with CD133+ BMDSCs for endometrial regeneration in Asherman’s syndrome

2021

Abstract Study question Can CD133+ bone marrow-derived stem cells (BMDSCs) loaded in polyethylene glycol diacrylate (PEGda) and gelatin divide and decidualize? Summary answer Biocompatible porous PEGda and gelatin scaffold provides a three-dimensional environment for CD133+ cells to attach, divide, and decidualize in vitro. What is known already Intrauterine adhesions (IUA) develop due to acquired damages in the endometrium resulting in partial to complete endometrial dysfunction in the Asherman syndrome. Previous works from our group have demonstrated the engraftment of CD133+ BMDSCs and its paracrine effect on endometrial proliferation, improved endometrial thickness and clinical outcome …

Sustainable Regeneration of Brownfields

2016

Our cities are really rich of building ensembles now deserted which were, in the recent past, sources of job and productivity for the community. They are generally inside the urban fabric or in the suburbs and they are often not particularly taken into consideration because they haven’t architectonic characteristics which could justify a restoration. Due to their big extension, the planning approach turns to be diffucult. Besides, these areas, if adequately restored, could become an important economic and cultural resource and a possibility of experimentation of new sustainable technologies. All this heritage requires a planning approach based on conservation and valorization of all the cul…

Disruption of the retinitis pigmentosa 28 gene Fam161a in mice affects photoreceptor ciliary structure and leads to progressive retinal degeneration.

2014

Mutations in the FAM161A gene were previously identified as the cause for autosomal-recessive retinitis pigmentosa 28. To study the effects of Fam161a dysfunction in vivo, we generated gene-trapped Fam161a(GT/GT) mice with a disruption of its C-terminal domain essential for protein-protein interactions. We confirmed the absence of the full-length Fam161a protein in the retina of Fam161a(GT/GT) mice using western blots and showed weak expression of a truncated Fam161a protein by immunohistochemistry. Histological analyses demonstrated that photoreceptor segments were disorganized in young Fam161a(GT/GT) mice and that the outer retina was completely lost at 6 months of age. Reactive microglia…
