Delivery of proteins into living cells by reversible membrane permeabilization with streptolysin-O

The pore-forming toxin streptolysin O (SLO) can be used to reversibly permeabilize adherent and nonadherent cells, allowing delivery of molecules with up to 100 kDa mass to the cytosol. Using FITC-labeled albumin, 10 5 –10 6 molecules were estimated to be entrapped per cell. Repair of toxin lesions depended on Ca 2+ -calmodulin and on intact microtubules, but was not sensitive to actin disruption or to inhibition of protein synthesis. Metabotropic glutamate receptors activate phospholipase D in astrocytes through a protein kinase C-dependent and Rho-independent pathway.

Metabotropic glutamate receptors (mGluRs) are G protein-coupled receptors that mediate phospholipase D (PLD) activation in brain, but the mechanism underlying this response remains unclear. Here we used primary cultures of astrocytes as a cell model to explore the mechanism that links mGluRs to PLD. Glutamate activated both phospholipase C (PLC) and PLD with equal potency and this effect was mimicked by L-cysteinesulfinic acid, a putative neurotransmitter previously shown to activate mGluRs coupled to PLD, but not PLC, in adult brain. Family Well-Being Under Pressure: Rhythmanalysis Applied to Post Pandemic Family Dynamics

This contribution deals with the centrality of family relationships for the production of individual well-being, both on a cognitive level, i.e. as an influence on satisfaction with one’s own life and, therefore, as the outcome of a cognitive evaluation, and on an experiential and emotional level, i.e. as an influence on moment-to-moment experience. It starts from an analysis of subjective well-being, arriving at a concept of relational well-being, whilst proposing an approach of rhythmanalysis as a tool for understanding the changes occurring in family dynamics in the post-Covid era.

The current study aimed to examine the nature of the coupling between olfactory stimuli from the mother and the newborn's crawling and rooting (exploratory movements of the head). Taxonomy is a knowledge management tool that presents useful information in a well-ordered structure prevents overloading of information on its access and making the information access qualitative. This article is concerned with automatically extracting asymmetrical hierarchical relations from a large corpus and subsequent taxonomy construction by domain independent and semi-supervised system.
