The assessment of genotoxic potential in surface water requires test methods, among which are those that detect initial DNA damage in organisms of aquatic biocenosis. The microgel electrophoresis (MGE) "comet assay" was applied to a ubiquitous unicellular green alga (Chlamydomonas reinhardtii) to detect DNA damage caused by genotoxins. For this, the test protocol described by Singh NP et al. [Exp Cell Res 175: 184-191, 1988] was modified. International audience; This paper reports the development of a highly sensitive analytical method combining solid-phase extraction (SPE) with ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UHPLC–MS/MS), for the monitoring of ultra-trace levels of N-nitrosamines in water samples. Under optimized analytical conditions, chromatographic separation was performed in 3 min, in isocratic mode, using an Acquity UHPLC C18 column and a mobile phase consisting of acetonitrile, water, and formic acid (60:40:0.1, v/v/v) at a flow rate of 0.4 mL min−1. The biological effect of putative genotoxic chemicals in the work place environment was monitored in peripheral mononuclear blood cells of exposed workers. DNA strand breaks, alkali-labile sites of DNA and DNA cross-links were measured using the alkaline filter elution method. A dose dependent increase in DNA damage was found in sterilization workers exposed to ethylene oxide and metal workers with exposure towards N-nitrosodiethanolamine. Two subpopulations with different response to the external exposure were found in nonsmoking sterilization workers. In order to describe the influence of Pleistocene glaciations on the genetic structure and demography of a highly mobile, but specialized, passerine, the Savi's Warbler (Locustella luscinioides), mitochondrial DNA sequences (ND2) and microsatellites were analysed in c.330 individuals of 17 breeding and two wintering populations. Phylogenetic, population genetics and coalescent methods were used to describe the genetic structure, determine the timing of the major splits and model the demography of populations.
