New Insights of Oral Colonic Drug Delivery Systems for Inflammatory Bowel Disease Therapy
2020
[EN] Colonic Drug Delivery Systems (CDDS) are especially advantageous for local treatment of inflammatory bowel diseases (IBD). Site-targeted drug release allows to obtain a high drug concentration in injured tissues and less systemic adverse effects, as consequence of less/null drug absorption in small intestine. This review focused on the reported contributions in the last four years to improve the effectiveness of treatments of inflammatory bowel diseases. The work concludes that there has been an increase in the development of CDDS in which pH, specific enzymes, reactive oxygen species (ROS), or a combination of all of these triggers the release. These delivery systems demonstrated a th…
Absence of endo-1,4-β-glucanase KOR1 alters the Jasmonate-dependent defence response to Pseudomonas syringae in Arabidopsis
2014
During plant-pathogen interactions, the plant cell wall forms part of active defence against invaders. In recent years, cell wall-editing enzymes, associated with growth and development, have been related to plant susceptibility or resistance. Our previous work identified a role for several tomato and Arabidopsis endo-1,4-β-glucanases (EGs) in plant-pathogen interactions. Here we studied the response of the Arabidopsis thaliana T-DNA insertion mutant lacking EG Korrigan1 (KOR1) infected with Pseudomonas syringae. KOR1 is predicted to be an EG which is thought to participate in cellulose biosynthesis. We found that kor1-1 plants were more susceptible to P. syringae, and displayed severe dise…
Induction of tcI 7 , a gene encoding a β-subunit of proteasome, in tobacco plants treated with elicitins, salicylic acid or hydrogen peroxide 1
2000
We previously isolated, by differential display and 5′ RACE (rapid amplification of cDNA ends), cDNAs corresponding to genes activated following cryptogein treatment of tobacco cell suspensions, among them tcI 7 (tcI for obacco ryptogein nduced), a gene encoding a β-subunit of proteasome. Here, we report that tcI 7 was up-regulated in tobacco plants treated with elicitins (cryptogein and parasiticein) that have been shown to induce a systemic acquired resistance (SAR). Moreover, subsequent inoculation of tobacco with the pathogen Phytophthora parasitica var. nicotianae (Ppn) was shown to induce an additional activation of tcI 7 in tobacco plants pretreated with cryptogein. We also showed an…
Evaluation of the pH effect of formulations on the skin permeability of drugs by biopartitioning micellar chromatography☆
2004
Dermal absorption of chemicals is an area of increasing interest for the pharmaceutical and cosmetic industries, as well as in dermal exposure and risk assessment processes. Biopartitioning micellar chromatography (BMC) is a mode of reversed phase micellar chromatography that has proved to be useful in the description and prediction of several pharmacological properties of xenobiotics including oral drug absorption, ocular and skin drug permeability. The present paper deals with the application of biopartitionig micellar chromatography to evaluate the pH effect on the skin permeability of twelve non-steroidal anti-inflammatory drugs and lidocaine. For this purpose the BMC retention of the w…
Hyaluronic Acid Increases Anti-Inflammatory Efficacy of Rectal 5-Amino Salicylic Acid Administration in a Murine Colitis Model
2021
5-amino salicylic acid (5-ASA) is a standard therapy for the treatment of mild to moderate forms of inflammatory bowel diseases (IBD) whereas more severe forms involve the use of steroids and immunosuppressive drugs. Hyaluronic acid (HA) is a naturally occurring non-sulfated glycosaminoglycan that has shown epithelium protective effects in experimental colitis recently. In this study, both 5-ASA (30 mg/kg) and HA (15 mg/kg or 30 mg/kg) were administered rectally and investigated for their potential complementary therapeutic effects in moderate or severe murine colitis models. Intrarectal treatment of moderate and severe colitis with 5-ASA alone or HA alone at a dose of 30 mg/kg led to a sig…
In vitro study of N-succinyl chitosan for targeted delivery of 5-aminosalicylic acid to colon
2011
In vitro study on N-succinyl chitosan and chitosan (control) matrices for selective colon delivery of 5-aminosalicylic acid is described. Matrices containing β-cyclodextrin were also prepared. As shown by DSC analyses, the drug was successfully loaded into the matrices reaching up to 95% entrapment efficiency. Swelling and drug release were studied at pH 1.2, pH 7.4, and in a pH gradient medium to simulate the gastro-intestinal transit. Main result of this study was a higher capability of N-succinyl chitosan alone to better control drug release in the simulated gastro-intestinal transit: N-succinyl chitosan gave the lowest release in acid medium (≅15%) and the highest in alkaline environmen…
Monetary valuation of salicylic acid, methylparaben and THCOOH in a Mediterranean coastal wetland through the shadow prices methodology
2018
The presence of pharmaceutical and personal care products and drugs of abuse (PPCPs) in wastewater treatment plants (WWTPs) indicates discharge of the effluent may not be suitable for the ecological balance of water ecosystems, such as wetlands. These PPCPs degrade water quality, considered as an ecosystem service (ES), provoking serious environmental impacts. Assessing the monetary value of PPCPs can be used as a proxy for environmental status of the ES of water quality (ESWQ). Considering PPCPs as non-desirable outputs of WWTPs, the shadow prices methodology has been implemented using directional distance function to measure the environmental avoided cost of removing salicylic acid (SA), …
Two new biologically active triterpenoidal saponins acylated with salicylic acid from Albizia adianthifolia
2003
International audience; Two new oleanane-type triterpene saponins, adianthifoliosides A (1) and B (2), were isolated from a 95% ethanolic extract of roots of Albizia adianthifolia. Their structures were elucidated mainly by using a combination of 600 MHz 1D and 2D NMR techniques (COSY, NOESY, TOCSY, HSQC, and HMBC) and by FABMS and HRESIMS. Compounds 1 and 2 were characterized as glycosides of acacic acid acylated by an o-hydroxybenzoyl unit. The crude saponin mixture (CSM), compounds 1 and 2 together with 3 and 4 (prosapogenins obtained from the mild alkaline hydrolysate of the CSM), were evaluated for immunomodulatory activity on the Jurkat T cell line and for hemolytic property against s…
Surface processes in selective photocatalytic oxidation of hydroxybenzyl alcohols by TiO2 P25
2023
Aryl aldehydes and acids are important industrial intermediates, but their synthesis from the corresponding alcohols often requires the use of high temperatures and harmful solvents and oxidant agents. Heterogeneous photocatalysis represents an interesting option for the partial oxidation of alcohols to aldehydes at room temperature using water as solvent. In this work, the partial photocatalytic oxidation of 2-hydroxybenzyl alcohol (salicyl alcohol) and 4-hydroxybenzyl alcohol to the corresponding aldehydes and acids has been compared, under environmental-friendly conditions, employing TiO2 P25 as photocatalyst in water. The selectivity to the corresponding aldehydes and acids, under the s…
Free radical-mediated systemic immunity in plants.
2014
SPE IPM; Systemic acquired resistance (SAR) is a form of defense that protects plants against a broad-spectrum of secondary infections by related or unrelated pathogens. SAR related research has witnessed considerable progress in recent years and a number of chemical signals and proteins contributing to SAR have been identified. All of these diverse constituents share their requirement for the phytohormone salicylic acid, an essential downstream component of the SAR pathway. However, recent work demonstrating the essential parallel functioning of nitric oxide (NO)-derived and reactive oxygen species (ROS)-derived signaling together with SA provides important new insights in the overlapping …
