The Peroxisomal 3-keto-acyl-CoA thiolase B Gene Expression Is under the Dual Control of PPARα and HNF4α in the Liver
2011
PPARα and HNF4α are nuclear receptors that control gene transcription by direct binding to specific nucleotide sequences. Using transgenic mice deficient for either PPARα or HNF4α, we show that the expression of the peroxisomal3-keto-acyl-CoA thiolase B(Thb) is under the dependence of these two transcription factors. Transactivation and gel shift experiments identified a novel PPAR response element within intron 3 of theThbgene, by which PPARα but not HNF4α transactivates. Intriguingly, we found that HNF4α enhanced PPARα/RXRα transactivation from TB PPRE3 in a DNA-binding independent manner. Coimmunoprecipitation assays supported the hypothesis that HNF4α was physically interacting with RXR…
Ultradeep Sequencing Analysis of Population Dynamics of Virus Escape Mutants in RNAi-Mediated Resistant Plants
2012
Plant artificial micro-RNAs (amiRs) have been engineered to target viral genomes and induce their degradation. However, the exceptional evolutionary plasticity of RNA viruses threatens the durability of the resistance conferred by these amiRs. It has recently been shown that viral populations not experiencing strong selective pressure from an antiviral amiR may already contain enough genetic variability in the target sequence to escape plant resistance in an almost deterministic manner. Furthermore, it has also been shown that viral populations exposed to subinhibitory concentrations of the antiviral amiR speed up this process. In this article, we have characterized the molecular evolutiona…

MOJAVE. XIII. PARSEC-SCALE AGN JET KINEMATICS ANALYSIS BASED ON 19 YEARS OF VLBA OBSERVATIONS AT 15 GHz
2016
We present 1625 new 15 GHz (2 cm) VLBA images of 295 jets associated with active galactic nuclei (AGNs) from the MOJAVE and 2 cm VLBA surveys, spanning observations between 1994 Aug 31 and 2013 Aug 20. For 274 AGNs with at least 5 VLBA epochs, we have analyzed the kinematics of 961 individual bright features in their parsec-scale jets. A total of 122 of these jets have not been previously analyzed by the MOJAVE program. In the case of 451 jet features that had at least 10 epochs, we also examined their kinematics for possible accelerations. At least half of the well-sampled features have non-radial and/or accelerating trajectories, indicating that non-ballistic motion is common in AGN jets.…
Causative role of oxidative stress in a Drosophila model of Friedreich ataxia
2006
Friedreich ataxia (FA), the most common form of hereditary ataxia, is caused by a deficit in the mitochondrial protein frataxin. While several hypotheses have been suggested, frataxin function is not well understood. Oxidative stress has been suggested to play a role in the pathophysiology of FA, but this view has been recently questioned, and its link to frataxin is unclear. Here, we report the use of RNA interference (RNAi) to suppress the Drosophila frataxin gene (fh) expression. This model system parallels the situation in FA patients, namely a moderate systemic reduction of frataxin levels compatible with normal embryonic development. Under these conditions, fh-RNAi flies showed a shor…
Evidence for the presence of functional protease activated receptor 4 (PAR4) in the rat colon
2004
Background and aims: Protease activated receptors (PARs) have been postulated to play a role during intestinal inflammation. The presence and role played by PAR4 in gastrointestinal functions have not been fully clarified. The aims of this study were: (i) to examine expression of PAR4 in rat proximal colon; (ii) to determine the mechanical effects induced by PAR4 activation in longitudinal muscle; and (iii) to characterise the underlying mechanisms. Methods: PAR4 expression was determined by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. Mechanical activity was recorded as changes in isometric tension. Results: A PCR product corresponding to the predicted…
Eye tracking analysis of minor details in films for audio description
2012
This article focuses on the many instances when minute details found in feature films may have direct implications upon the development of both the visual and plot narratives. The main question we would like to ask examines whether very subtle details which may easily go unnoticed by the viewer should be audio described. To assess the visual consciousness of such minute details, a perception experiment was conducted using eye-tracking technology and questionnaires. Though the result is not conclusive, it shows a clear methodological approach in the field of the audio description of visual details, and does give some indication as to what should be taken into consideration in future studies …
Multi-sensory approaches to (audio) describing the visual arts
2012
Making art accessible to blind patrons requires the ability to convey explicit and implicit visual messages through non-visual forms. Audio description is often seen as the best way to offer visual texts to blind people; however, one may query whether words alone are sufficient to convey the subtleties of art and to transport the emotional charge such works offer. It is a fact that the dialogue between words and touch may allow these particular “readers” to “see” art in its tangible forms, but perhaps more is needed if one is to give them the opportunity to live the art experience to the full. This article discusses different multi-sensory approaches to making art available to visually impa…
Lyrics against images: music and audio description
2012
Music has been an integral part of films from as far back as the silent movie era, where a piano accompaniment was used to build the narrative and an orchestra was used to drown out the sound of the projector. Music in films can encompass a wide array of elements, from a song to a whistle, and can play many roles, as has been commented upon by many researchers. Due to the relevance of music in film narrative, it is an element which merits attention when it comes to making a film accessible, be it through Audio description, Audio subtitling, Sign Language or Subtitling for the Deaf and Hard-of-Hearing. This article focuses on Audio description (AD) and how music is handled in audio described…
