In Candida parapsilosis the ATC1 Gene Encodes for an Acid Trehalase Involved in Trehalose Hydrolysis, Stress Resistance and Virulence

2014

An ORF named CPAR2-208980 on contig 005809 was identified by screening a Candida parapsilosis genome data base. Its 67% identity with the acid trehalase sequence from C. albicans (ATC1) led us to designate it CpATC1. Homozygous mutants that lack acid trehalase activity were constructed by gene disruption at the two CpATC1 chromosomal alleles. Phenotypic characterization showed that atc1Δ null cells were unable to grow on exogenous trehalose as carbon source, and also displayed higher resistance to environmental challenges, such as saline exposure (1.2 M NaCl), heat shock (42°C) and both mild and severe oxidative stress (5 and 50 mM H2O2). Significant amounts of intracellular trehalose were …

Homozygous deletion of ATC1 and NTC1 genes in Candida parapsilosis abolishes trehalase activity and affects cell growth, sugar metabolism, stress res…

2015

A double homozygous atc1Δ/atc1Δ/ntc1Δ/ntc1Δ mutant (atc1Δ/ntc1Δ KO) was constructed in the pathogen opportunistic yeast Candida parapsilosis by disruption of the two chromosomal alleles coding for NTC1 gene (encoding a neutral trehalase) in a Cpatc1Δ/atc1Δ background (atc1Δ KO strain, deficient in acid trehalase). The Cpatc1Δ/ntc1Δ KO mutant failed to counteract the inability of Cpatc1Δ cells to metabolize exogenous trehalose and showed a similar growth pattern on several monosaccharides and disaccharides. However, upon prolonged incubation in either rich medium (YPD) or nutrient-starved medium the viability of Cpatc1Δ cells exhibited a sensitive phenotype, which was augmented by further Cp…

New approach for synthesis of poly(ethylglyoxylate) using Maghnite-H + , an Algerian proton exchanged montmorillonite clay, as an eco-catalyst

2017

International audience; In this works, we have explored a new method for a green synthesis of poly(ethylglyoxylate) (PEtG). This method consists on using a montmorillonite clay called Maghnite-H+ as an eco-catalyst to replace triethylamine which is toxic. Cationic polymerization experiments are performed in bulk conditions at three temperatures (-40 degrees C, 25 degrees C, 80 degrees C) and in THF solutions at room temperature (25 degrees C). At 25 degrees C, an optimum ratio of 5 wt% of catalyst leads to molar masses up to 22000 g/mol in THF solutions. Polymerizations in bulk conditions lead to slightly lower masses than experiments conducted in THF solutions. However, bulk polymerization…

Translation of hepatitis B virus (HBV) surface proteins from the HBV pregenome and precore RNAs in Semliki Forest virus-driven expression.

2004

Hepatitis B virus (HBV) pregenome RNA (pgRNA) serves as a translation template for the HBV core (HBc) protein and viral polymerase (Pol). HBV precore RNA (pcRNA) directs the synthesis of the precore (preC) protein, a precursor of the hepatitis B e antigen (HBeAg). pgRNA and pcRNA were expressed in the Semliki Forest virus (SFV) expression system. Besides the HBc and preC proteins, there was revealed the synthesis of all three forms of HBV surface (HBs) proteins: long (LHBs), middle (MHBs) and short (SHBs), the start codons of which are located more than 1000 nt downstream of the HBc and preC start codons. Moreover, other HBV templates, such as 3′-truncated pgRNA lacking 3′ direct repeat and…

Assays for Detection of Fungal Hallucinogens Such as Psilocybin and Psilocin

2016

Abstract Methods applicable to the detection of hallucinogenic indole compounds, namely psilocybin, psilocin, and related molecules, that have been developed so far for purposes of mycological research, toxicological studies, or forensic investigations are described in this chapter. The most current literature as well as some interesting older publications are cited, providing a historical overview of the subject. Methods of qualitative and quantitative analysis are presented. Additionally, preliminary purification steps and sample treatment before the exact quantitative determination are proposed. The methods described utilized various analytical techniques for the measurement of psilocybi…

Fungal Hallucinogens Psilocin, Ibotenic Acid, and Muscimol

2013

Psychoactive drugs of fungal origin, psilocin, ibotenic acid, and muscimol among them have been proposed for recreational use and popularized since the 1960s, XX century. Despite their well-documented neurotoxicity, they reached reputation of being safe and nonaddictive. Scientific efforts to find any medical application for these hallucinogens in psychiatry, psychotherapy, and even for religious rituals support are highly controversial. Even if they show any healing potential, their usage in psychotherapy is in some cases inadequate and may additionally harm seriously suffering patients. Hallucinogens are thought to reduce cognitive functions. However, in case of indolealkylamines, such as…

Atmospheric pressure plasma polymerisation of metalloporphyrins containing mesoporous membranes for gas sensing applications

2013

Abstract Metalloporphyrins are embedded in an organosilicon matrix by an easily up-scalable atmospheric pressure dielectric barrier discharge method. The integrity of the metalloporphyrins, followed by UV–visible spectroscopy, is successfully preserved and their aggregation prevented. The single molecule properties, rather than the bulk ones, are thus enhanced. Exposure to triethylamine, which reaches the metalloporphyrins through the pores of the organosilicon membrane, led to a shift in the absorption spectrum and confirms the gas sensing potential of such coatings.
