Zinc Binding Sites Conserved in Short Neuropeptides Containing a Diphenylalanine Motif

A diphenylalanine motif in peptides plays a crucial role in supramolecular systems. The current work represents a novel strategy in which a diphenylalanine motif in the central domain of neuropeptides conserves the specific Zn2+ binding site and prevents "hopping" of the Zn2+ ion between alternative metal binding sites. Alternative metal binding sites may also include carboxylic atoms in the terminal domains of a peptide. Therefore, one needs to design a peptide in which the metal will not bind the carboxylic groups in the terminal domains. Herein, we propose that engineering and designing peptides with a diphenylalanine motif in the central domain may yield excellent metal chelators.

Hemoglobin from the leech Macrobdella decora belongs to the class of giant extracellular hexagonal bilayer globin structures found in annelid and vestimentiferan worms. These complexes consist of 144 heme-bearing subunits, exhibit a characteristic quaternary structure (2 × (6 × (3 × 4))), and contain tetramers as basic substructures that express cooperative oxygen binding and thus provide a structural basis for a hierarchy in allosteric interactions. Abstract The kinetics of the oxidation of malonic acid and (its substituted compounds, methyl-, ethyl-, butyl- and benzyl-malonic acid) by ceric ions has been studied at 20.0 °C in the absence and the presence of the surfactant N-tetradecyl-N,N-dimethylamine oxide (C 14 DMAO). The addition of increasing amounts of C 14 DMAO influences the rate of the redox process to an extent that significantly depends on the hydrophobicity of the reducing species. Four new complexes, [Cu2(C2O4) 2(SCS)]·3H2O (1), [Cu2(C 2O4)3(SCSH)2]·H2O (2), [Cu(C2O4)(SS)2]·3H2O (3) and [Cu(C2O4)(SS)]·6H2O (4) [where SCS = bis(4-pyridylthio)methane and SS = 4,4′-dipyridyldisulfide], have been synthesized by the reaction of the SS and SCS ligands with the corresponding copper(ii) oxalate salt. Abstract The enantioselective epoxide biohydrolysis of the racemic benzodioxole 6,7-epoxygeraniol derivative 1 has been achieved using the fungus A. niger . This new type of preparative scale bioconversion allows the synthesis of both enantiomers of Bower's compound, an analogue of insect juvenile hormone. Biological tests showed that the 6(R) enantiomer was about ten times more active than the 6(S) enantiomer against the yellow meal worm Tenebrio molitor .

Novel heterobimetallic radiotheranostic: preparation, activity, and biodistribution.

A novel Ru(II) (arene) theranostic complex is presented. It is based on a 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid macrocycle bearing a triarylphosphine and can be tracked in vivo by using the γ emission of (153) Sm atoms. Notably, the heteroditopic ligand can be selectively metalated with ruthenium at the phosphorus atom despite the presence of other functionalities that are prone to metal coordination. Subsequent labeling with radionuclides such as (153) Sm can then be performed easily. The resulting heterobimetallic complex exhibits favorable solubility and stability properties in biologically relevant media. It also shows in vitro cytotoxicity in line with that expected …
