Light microscopic data on the innervation of taste buds in the soft palate of the rat obtained by methylene blue (MB) supravital staining are presented here for the first time. The taste pore including subpopulations of taste bud cells and nerve fibers were stained. Intragemmal nerve fibers were detected ascending up to the taste pore; this suggests the possibility of additional paracellular pathways in taste transduction directly via nerve fibers. Moreover, relatively large bulbous or laminar enlargements of the intragemmal nerve fibers were found. The branches of these nerve fibers, showing minute spiny protuberances, appeared to anastomose. The taste buds in the soft palate and the circumvallate papillae of the rat were investigated by the zinc iodide-osmium tetroxide technique. In addition, electron micrographs of taste buds stained with this method were presented for the first time. Differences in taste bud structures were found between the examined regions. The taste buds of the soft palate showed a complicated plexus of intragemmal nerve fibers. Some fibers exhibited terminal polymorphic swellings. Single branches could be traced close to the space of the taste pore. Over the last decade, converging data have been accumulated both in rodents and humans, supporting the existence of a sixth taste modality devoted to the perception of dietary lipids. It is well known that the sense of taste is determinant for the food choice and that the overconsumption of highly palatable energy-dense foods contributes to the current obesity epidemic. Thus, an important issue in terms of Public Health is to understand the mechanisms by which the oro-sensory perception of fat is regulated. An overview of our current knowledge in this field of investigations is proposed in this mini-review.
