Validation of a high-performance chromatographic method for determination of cefotaxime in biological samples

An analytical method for detecting and quantifying cefotaxime in plasma and several tissues is described. The method was developed and validated using plasma and tissues of rats. The samples were analyzed by reversed phase liquid chromatography (HPLC) with UV detection (254 nm). Calibration graphs showed a linear correlation (r > 0.999) over the concentration ranges of 0.5–200 μg/mL and 1.25–25 μg/g for plasma and tissues, respectively. The recovery of cefotaxime from plasma standards prepared at the concentrations of 25 μg/mL and 100 μg/mL was 98.5 ± 3.5% and 101.8 ± 2.2%, respectively. Enzymatic water extraction of taxifolin from wood sawdust of Larix gmelini (Rupr.) Rupr. and evaluation of its antioxidant activity

Abstract An enzyme incubation–water extraction (EI–WE) method was developed and optimised for the extraction of the natural antioxidant taxifolin and of the total flavonoids from wood sawdust of Larix gmelini ( Rupr. ) Rupr. A factorial design and a central composite design approach were used for method optimisation. Optimal conditions were 0.5 mg/ml cellulase and 0.5 mg/ml pectinase, a pH of 5.0, a temperature of 32 °C and 18 h incubation time. The flavonoids and taxifolin were extracted in hot water at 50 °C for 30 min, with a solid to liquid ratio of 1:20. A Simple Technique for Measuring Leucocyte Chemotaxis in Reversible Boyden Chambers

A technique for assay of leucocyte chemotaxis in Boyden chambers is described. A set of up to 12 closed chambers with 2 compartments is formed from commercially available migration plates. The set can be turned during the incubation and thus the detachment of cells from the filter, a major source of variation in classical type of chambers, can be prevented. The technique is simple and so inexpensive that it allows the use of the material as disposable if preferred.

Serotoninergic Modulation of Phototactic Variability Underpins a Bet-Hedging Strategy in Drosophila melanogaster

When organisms’ environmental conditions vary unpredictably in time, it can be advantageous for individuals to hedge their phenotypic bets. It has been shown that a bet-hedging strategy possibly underlies the high inter-individual diversity of phototactic choice in Drosophila melanogaster. This study shows that fruit flies from a population living in a boreal and relatively unpredictable climate have more variable variable phototactic biases than fruit flies from a more stable tropical climate, consistent with bet-hedging theory. Spontaneous Spatiotemporal Ordering of Shape Oscillations Enhances Cell Migration

The migration of cells is relevant for processes such as morphogenesis, wound healing, and invasion of cancer cells. In order to move, single cells deform cyclically. However, it is not understood how these shape oscillations influence collective properties. Here we demonstrate, using numerical simulations, that the interplay of directed motion, shape oscillations, and excluded volume enables cells to locally "synchronize" their motion and thus enhance collective migration. Chemotaxis and Haptotaxis on Cellular Level

Chemotaxis and haptotaxis have been a main theme in the macroscopic study of bacterial and cellular motility. In this work, we use a successful model that describes cellular motility and investigate the influence these processes have on the shape and motility of fast migrating cells. We note that, despite the biological and modelling differences of chemotaxis and haptotaxis, the cells exhibit many similarities in their migration. In particular, after an initial adjustment phase, the cells obtain a stable shape, similar in both cases, and move with constant velocity.

Pattern formation and transition to chaos in a chemotaxis model of acute inflammation

We investigate a reaction-diffusion-chemotaxis system that describes the immune response during an inflammatory attack. The model is a modification of the system proposed in Penner, Ermentrout, and Swigon [SIAM J. Appl. Dyn. Syst., 11 (2012), pp. 629-660]. We introduce a logistic term in the immune cell dynamics to reproduce the macrophages' activation, allowing us to describe the disease evolution from the early stages to the acute phase.
