A headlight on liquid biopsies: a challenging tool for breast cancer management

Breast cancer is the most frequent carcinoma and second most common cause of cancer-related mortality in postmenopausal women. The acquisition of somatic mutations represents the main mechanism through which cancer cells overcome physiological cellular signaling pathways (e.g., PI3K/Akt/mTOR, PTEN, TP53). To date, diagnosis and metastasis monitoring is mainly carried out through tissue biopsy and/or re-biopsy, a very invasive procedure limited only to certain locations and not always feasible in clinical practice. Exploring the readthrough of nonsense mutations by non-acidic Ataluren analogues selected by ligand-based virtual screening

Abstract Ataluren, also known as PTC124, is a 5-(fluorophenyl)-1,2,4-oxadiazolyl-benzoic acid suggested to suppress nonsense mutations by readthrough of premature stop codons in the mRNA. Potential interaction of PTC124 with mRNA has been recently studied by molecular dynamics simulations highlighting the importance of H-bonding and stacking π−π interactions. A series of non-acidic analogues of PTC124 were selected from a large database via a ligand-based virtual screening approach. Detection of MET Alterations Using Cell Free DNA and Circulating Tumor Cells from Cancer Patients

MET alterations may provide a potential biomarker to evaluate patients who will benefit from treatment with MET inhibitors. Therefore, the purpose of the present study is to investigate the utility of a liquid biopsy-based strategy to assess MET alterations in cancer patients. We analyzed MET amplification in circulating free DNA (cfDNA) from 174 patients with cancer and 49 healthy controls and demonstrated the accuracy of the analysis to detect its alteration in patients. Abstract Post-Greimassian semiotics has worked toward a return to phenomenology, with the aim of studying the sensorial dimension and the body. Most of this research, however, has all but forgotten Greimas’s last book, De imperfection (1987), in which he proposes an original version of Merleau-Ponty’s philosophy with the notion of the aesthetic grasp. I propose to reconsider this almost abandoned notion, both from a theoretical point of view (a new version of the catharsis of philosophical aesthetics) and from the vantage of textual descriptions (the short story “The Naked Bosom” by Italo Calvino and the movie Ratatouille).

Enhancement of premature stop codon readthrough in the CFTR gene by Ataluren (PTC124) derivatives.

Abstract Premature stop codons are the result of nonsense mutations occurring within the coding sequence of a gene. These mutations lead to the synthesis of a truncated protein and are responsible for several genetic diseases. A potential pharmacological approach to treat these diseases is to promote the translational readthrough of premature stop codons by small molecules aiming to restore the full-length protein. Simple algorithms for calculation of the axial‐symmetric heat transport problem in a cylinder

The approximation of axial‐symmetric heat transport problem in a cylinder is based on the finite volume method. In the classical formulation of the finite volume method it is assumed that the flux terms in the control volume are approximated with the finite difference expressions. Then in the 1‐D case the corresponding finite difference scheme for the given source function is not exact. There we propose the exact difference scheme. In 2‐D case the corresponding integrals are approximated using different quadrature formulae. A fully adaptive wavelet algorithm for parabolic partial differential equations

We present a fully adaptive numerical scheme for the resolution of parabolic equations. It is based on wavelet approximations of functions and operators. Following the numerical analysis in the case of linear equations, we derive a numerical algorithm essentially based on convolution operators that can be efficiently implemented as soon as a natural condition on the space of approximation is satisfied. The algorithm is extended to semi-linear equations with time dependent (adapted) spaces of approximation. Numerical experiments deal with the heat equation as well as the Burgers equation.

Diffusion front capturing schemes for a class of Fokker–Planck equations: Application to the relativistic heat equation

In this research work we introduce and analyze an explicit conservative finite difference scheme to approximate the solution of initial-boundary value problems for a class of limited diffusion Fokker-Planck equations under homogeneous Neumann boundary conditions. We show stability and positivity preserving property under a Courant-Friedrichs-Lewy parabolic time step restriction. We focus on the relativistic heat equation as a model problem of the mentioned limited diffusion Fokker-Planck equations. Convergence of a high-order compact finite difference scheme for a nonlinear Black–Scholes equation

A high-order compact finite difference scheme for a fully nonlinear parabolic differential equation is analyzed. The equation arises in the modeling of option prices in financial markets with transaction costs. It is shown that the finite difference solution converges locally uniformly to the unique viscosity solution of the continuous equation. The proof is based on a careful study of the discretization matrices and on an abstract convergence result due to Barles and Souganides.
