Structure and properties of hydroxyl radical modified nucleic acid components II. 8-Oxo-adenine and 8-oxo-2′-deoxy-adenosine

Abstract The tautomerism of the 8-oxo-adenine (8-oxo-A) and 8-oxo-2′-deoxy-adenosine (8-oxo-dA) was analysed on the basis of semiempirical, SCF ab initio and DFT density functional quantum chemistry calculations. The results of full gradient geometry optimisation of all possible 8-oxo-A and 8-oxo-dA structures lead to the conclusion that the most stable form is 8-keto-6-amino-tautomer. The second stable tautomer corresponds to 8-hydroxy-isomer. Such an order was unchanged after solvating process. In all studied solvents: water, methanol, acetone, cyclohexane the keto-tautomer proceeds the enol one. The preferred N-glycoside torsion angle corresponds to syn rotamer of 8-oxo-dA in all studied…

CD surface molecules mediates cell activation and signaling. In particular, CD14 on blood monocytes mediate monocyte/macrophage activation by lipopolysaccharide. Lipopolysaccharide and its receptor, CD14, have been implicated in atherogenesis. It has been recently shown that a C(-260)T polymorphism in the promoter of the CD14 receptor may be a risk factor for coronary artery disease. Recently this association has been questioned because no increased risk was found with the T allele, even in the homozygous state. Cyclobutane Pyrimidine Photodimerization of DNA/RNA Nucleobases in the Triplet State

The photoinduced formation of cyclobutane pyrimidine dimers in the triplet excited state of the DNA/RNA pyrimidine nucleobases pairs has been studied at the CASPT2 level of theory. A stepwise mechanism through the triplet state of the homodimer is proposed for the pairs of nucleobases cytosine, thymine, and uracil involving a singlet−triplet crossing intermediary structure of biradical character representing the most favorable triplet state conformation of the nucleobases as found in the DNA environment. Aiming to find novel genes that contribute to the stability of the genome, we screened the Saccharomyces cerevisiae deletion collection for increased mutator phenotypes. Objective— Apolipoprotein A5 ( APOA5 ) is a key determinant of plasma triglyceride (TG) concentrations. Genetic variation at the APOA5 locus could be responsible for some of the observed differences in response to fenofibrate therapy. Methods and Results— We examined the association between tag SNPs (−1131T>C and 56C>G) at APOA5 and TG and HDL-C response to fenofibrate and a postprandial lipid challenge in 791 men and women participating in the GOLDN study. Trifluridine/tipiracil is an oral chemotherapeutic agent recently approved for third-line treatment of chemorefractory metastatic colorectal cancer. Isolation and Characterization of Epidermal DNA and RNA from Guinea Pig Skin

DNA and RNA were isolated from mammalian epidermis in a relatively small scale procedure. The high purity and native state of the DNA isolated is reflected by its molar absorptivity E (P), its thermal hyperchromicity and its hyperchromicity upon DNase treatment and by its sedimentation profile as well as by its profile in a cesium chloride density gradient. The very low content of protein and RNA, as well as the data of DNA determination, indicate that this method permits the isolation of a highly purified product.
