Factors that predict response of patients with hepatitis C virus infection to boceprevir
2012
Background & Aims Little is known about factors associated with a sustained virologic response (SVR) among patients with hepatitis C virus (HCV) infection to treatment with protease inhibitors. Methods Previously untreated patients (from the Serine Protease Inhibitor Therapy 2 [SPRINT-2] trial) and those who did not respond to prior therapy (from the Retreatment with HCV Serine Protease Inhibitor Boceprevir and PegIntron/Rebetol 2 [RESPOND-2] trial) received either a combination of peginterferon and ribavirin for 48 weeks or boceprevir, peginterferon, and ribavirin (triple therapy) after 4 weeks of peginterferon and ribavirin (total treatment duration, 28–48 wk). A good response to interfer…
Characteristics of patients with hepatitis C virus-related chronic liver diseases just before the era of oral direct-acting antiviral therapy in Italy
2018
Background In 2017, oral direct-acting antiviral (DAA) therapy for chronic hepatitis C virus (HCV) infection became available free of charge for all HCV-RNA-positive patients, irrespective of their fibrosis stage. Aim The aim of this study was to evaluate the characteristics of HCV-related chronic liver disease (CLD) in Italy just before the introduction of DAA therapy. Patients and methods Patients with CLD were enrolled in two national surveys conducted in 2001 and in 2014. The two surveys prospectively enrolled patients aged older than 18 years referring to Italian liver units throughout the country using a similar clinical approach and analytical methods. Results Out of the 12 564 patie…
Real-world outcomes in patients with chronic hepatitis C: primary results of the PROBE study.
2014
BACKGROUND This large prospective multicentre cohort study aimed to improve knowledge of therapy for chronic hepatitis C (CHC) in real clinical practice. METHODS A diverse population of adults with CHC including patients with comorbid conditions, laboratory abnormalities and demographic features [comorbidities or special populations (CSP)] who were under-represented or excluded from peginterferon registration studies was treated with peginterferon α-2a (40 kDa) or α-2b (12 kDa) plus ribavirin at the investigator's discretion. RESULTS During the study, 5399 treatment-naive patients [2527 (46.8%) with CSP] received peginterferon α-2a (n=3513, 65.1%) or peginterferon α-2b (n=1886, 34.9%). The …
The Impact of World and European Football Cups on Stroke in the Population of Dijon, France: A Longitudinal Study from 1986 to 2006
2013
Background: Acute stress may trigger vascular events. We aimed to investigate whether important football competitions involving the French football team increased the occurrence of stroke. Methods: We retrospectively retrieved data of fatal and nonfatal stroke during 4 World Football Cups (1986, 1998, 2002, and 2006) and 4 European Championships (1992, 1996, 2000, and 2004), based on data from the population-based Stroke Registry of Dijon, France. One period of exposure was analyzed: the period of competition extended to 15 days before and after the competitions. The number of strokes was compared between exposed and unexposed corresponding periods of preceding and following years using Poi…
Processing of word stress related acoustic information : A multi-feature MMN study
2016
In the present study, we investigated the processing of word stress related acoustic features in a word context. In a passive oddball multi-feature MMN experiment, we presented a disyllabic pseudo-word with two acoustically similar syllables as standard stimulus, and five contrasting deviants that differed from the standard in that they were either stressed on the first syllable or contained a vowel change. Stress was realized by an increase of f0, intensity, vowel duration or consonant duration. The vowel change was used to investigate if phonemic and prosodic changes elicit different MMN components. As a control condition, we presented non-speech counterparts of the speech stimuli. Result…
Evaluation of intraindividual contrast enhancement variability for determining the maximum achievable consistency in CT
2019
OBJECTIVE. The purpose of this study was to quantify temporal variability in vascular and parenchymal enhancement within the same patient and to determine technique-related factors contributing to this variability. MATERIALS AND METHODS. We identified 100 patients who underwent four CT scans within 12 months with identical acquisition and contrast injection parameters. Enhancement was recorded in the abdominal aorta, main portal vein, liver parenchyma, and subcutaneous fat. Patient demographic and body habitus data were recorded. Injection-related factors were recorded including delay time from contrast injection to image acquisition. All pairwise differences in enhancement within each pati…
Influence of intracellular convection on the oxygen release by human erythrocytes
1972
There is general agreement today that intracellular diffusive transport of HbO2 and O2 limits the rate of oxygen uptake or release by the blood in the exchange vessels. Recent hemorheological results have shown that the mammalian erythrocyte exhibits fluidity as its most unique rheological property: it can be deformed continuously and rapidly, shear and normal stresses can be transmitted to the interior of the cell where systems of laminar flow are induced. These mechanical properties lead to the question whether or not intracellular convection does take place in the erythrocyte and to what extent it plays a part in gas exchange. A method was developed which subjects oxygen-saturated soluti…
Guideline-adherence regarding critical time intervals in the German Chest Pain Unit registry
2020
Background: Since 2008, the German Cardiac Society certified 256 Chest Pain Units (CPUs). Little is known about adherence to recommended performance measures in patients with suspected acute coronary syndrome (ACS) presenting to CPUs. We investigated guideline-adherence regarding critical time intervals and selected performance measures in German Chest Pain Units. Methods: From 2008 to 2014, 23,804 consecutive patients with suspected ACS were prospectively enrolled in the Chest Pain Unit registry of the German Cardiac Society. Results: Median time from symptom onset to first medical contact was 2 h in patients with ST-elevation myocardial infarction (STEMI) and 4 h in patients with unstable…
Delayed post-ischemic administration of CDP-choline increases EAAT2 association to lipid rafts and affords neuroprotection in experimental stroke
2007
Glutamate transport is the only mechanism for maintaining extracellular glutamate concentrations below excitotoxic levels. Among glutamate transporters, EAAT2 is responsible for up to 90% of all glutamate transport and has been reported to be associated to lipid rafts. In this context, we have recently shown that CDP-choline induces EAAT2 translocation to the membrane. Since CDP-choline preserves membrane stability by recovering levels of sphingomyelin, a glycosphingolipid present in lipid rafts, we have decided to investigate whether CDP-choline increases association of EAAT2 transporter to lipid rafts. Flotillin-1 was used as a marker of lipid rafts due to its known association to these m…
Downregulation and Nuclear Relocation of MLP During the Progression of Right Ventricular Hypertrophy Induced by Chronic Pressure Overload
2000
Abstract The cardiac LIM domain protein MLP plays a crucial role in the architecture and mechanical function of cardiac myocytes. Mice lacking the MLP gene develop cardiac hypertrophy, dilated cardiopathy and heart failure. We investigated whether downregulation of MLP is induced by pressure overload and contributes to the physiopathology of cardiac hypertrophy and failure. We studied this mechanism in rat right ventricles submitted to pulmonary arterial hypertension, because it is known that this ventricle is very vulnerable to the deleterious effects of pressure overload. During the progression of cardiac hypertrophy to failure over a 31 days period there was a dramatic decrease by 50% of…
