In the brain and in the plasma of patients with Alzheimer’s disease (AD), marked accumulation of C22:0 and of very long chain fatty acids (C24:0 ; C26:0) have been reported. Important decreases of docosahexaenoic acid (DHA; C22:6 n-3) have also been described as well as quantitative and qualitative modifications of plasmalogens. Altogether, these lipid modifications suggest an implication of peroxisomal metabolism disorders in the physiopathology of AD. The synthesis of secondary phosphine borane amino acids or dipeptides and their applications for the preparation of chiral ligands or for the grafting of fullerene, is described. These compounds were synthesized in good yield (up to 98%) without racemization. The principle of the synthesis is based on the alkylation of primary phosphine borane with a γ-iodo amino acid using phase transfer conditions. Tertiary diphosphine amino acids are obtained with 70% yield after a second alkylation. The aim of this PhD thesis is to elaborate mesoporous silica nanoparticles (MSNs) able to sustain the release of cisplatin into the intracellular compartments of solid tumors. The parametric study shows that morphological, structural and textural properties of MSNs-MCM-41, synthesized by sol-gel reaction by using TEOS as a silica source and CTAB as a structure-directing agent, depend on pH, stirring speed, temperature and extraction process of CTAB.
