Oligosaccharide recognition by selectins: Synthesis and biological activity of multivalent sialyl lewis-X ligands

Abstract Trivalent sialyl Lewis-X ligands 6–8 anchored onto flexible templates have been synthesized and evaluated as inhibitors of E-selectin and P-selectin mediated cell adhesion in cell culture assays and in vivo. Biological activities in vitro correlated with spacer length and lead to ligands with 3-fold (E-selectin) and 5-fold (P-selectin) improved receptor binding avidity per single tetrasaccharide moiety.

Molecular Design of the Cαβ Interface Favors Specific Pairing of Introduced TCRαβ in Human T Cells

Abstract A promising approach to adoptive transfer therapy of tumors is to reprogram autologous T lymphocytes by TCR gene transfer of defined Ag specificity. An obstacle, however, is the undesired pairing of introduced TCRα- and TCRβ-chains with the endogenous TCR chains. These events vary depending on the individual endogenous TCR and they not only may reduce the levels of cell surface-introduced TCR but also may generate hybrid TCR with unknown Ag specificities. We show that such hybrid heterodimers can be generated even by the pairing of human and mouse TCRα- and TCRβ-chains. Layer-by-layer deposited multilayer assemblies of polyelectrolytes and proteins: from ultrathin films to protein arrays

We have recently introduced a new method of creating ultrathin films of polyelectrolytes based on the electrostatic attraction between opposite charges. Multilayer assemblies are adsorbed in a layer-by-layer fashion from aqueous solutions of the polymers. The total film thickness can easily be adjusted by varying the ionic strength of the solution. Here, we report on the temperature stability and the water content of the multilayer assemblies. Furthermore, we have extended our concept to the incorporation of protein layers into films of synthetic polyelectrolytes. In this thesis DNA based structures were utilized to create gold nanostructures for nanosensing and nanoelectronic applications.
