Vemurafenib Inhibits Acute and Chronic Enterovirus Infection by Affecting Cellular Kinase Phosphatidylinositol 4-Kinase Type IIIβ
2023
Enteroviruses are one of the most abundant viruses causing mild to serious acute infections in humans and also contributing to chronic diseases like type 1 diabetes. Presently, there are no approved antiviral drugs against enteroviruses. Here, we studied the potency of vemurafenib, an FDA-approved RAF kinase inhibitor for treating BRAFV600E mutant-related melanoma, as an antiviral against enteroviruses. We showed that vemurafenib prevented enterovirus translation and replication at low micromolar dosage in an RAF/MEK/ERK-independent manner. Vemurafenib was effective against group A, B, and C enteroviruses, as well as rhinovirus, but not parechovirus or more remote viruses such as Semliki Fo…
Human Enterovirus Group B Viruses Rely on Vimentin Dynamics for Efficient Processing of Viral Nonstructural Proteins.
2019
A virus needs the host cell in order to replicate and produce new progeny viruses. For this, the virus takes over the host cell and modifies it to become a factory for viral proteins. Irrespective of the specific virus family, these proteins can be divided into structural and nonstructural proteins. Structural proteins are the building blocks for the new progeny virions, whereas the nonstructural proteins orchestrate the takeover of the host cell and its functions. Here, we have shown a mechanism that viruses exploit in order to regulate the host cell. We show that viral protein synthesis induces vimentin cages, which promote production of specific viral proteins that eventually control apo…
Informal learning in youth volunteer work : perspectives from the European Voluntary Service programme
2013
The purpose of this study is to examine the link between volunteer work and informal learning among young people. Drawing on the concepts and theories of lifelong learning, informal learning and volunteer work, and building on past research, this thesis explores the case of the European Voluntary Service (EVS), an international volunteering scheme devised by European Commission for youth between 18 and 30 years of age. Through 10 qualitative interviews with former and current EVS volunteers, this study attempts to understand the nature and main features of informal learning in EVS as well as what young people believe they have learnt in the course of their volunteering. The results show tha…
Viral Tweets, Fake News and Social Bots in Post-Factual Politics
2018
•PurposeIn the wake of Brexit and the 2016 US Presidential Elections, “post-factual” society has been heralded as a new era of political communications, where the digital public sphere plays a central role, in spreading “viral” contents and “fake news”, with the help of automated accounts or “social bots”. This paper seeks to define these terms and the methods by which the phenomena they commonly designate might be studied, in order to characterise the dynamics of political deliberation during the 2017 French Presidential Elections on Twitter, the online platform most commonly used for political communication in France. It thus aims to better understand the mechanisms by which information b…
Machine learning for mortality analysis in patients with COVID-19
2020
This paper analyzes a sample of patients hospitalized with COVID-19 in the region of Madrid (Spain). Survival analysis, logistic regression, and machine learning techniques (both supervised and unsupervised) are applied to carry out the analysis where the endpoint variable is the reason for hospital discharge (home or deceased). The different methods applied show the importance of variables such as age, O2 saturation at Emergency Rooms (ER), and whether the patient comes from a nursing home. In addition, biclustering is used to globally analyze the patient-drug dataset, extracting segments of patients. We highlight the validity of the classifiers developed to predict the mortality, reaching…
Membrane Protein Complexes: Structure and Function
2018
This edited book contains a compilation of 14 advanced academic chapters dealing with the structure and function of membrane protein complexes. This rapidly advancing important field of study closely parallels those on soluble protein complexes, and viral protein and nucleoprotein complexes.Diverse topics are included in this book, ranging from membrane–bound enzymes to ion channels, proton pumps and photosystems. Data from X-ray crystallography, cryo-electron microscopy and other biophysical and biochemical techniques are presented throughout the book. There is extensive use of colour figures of protein structures. Throughout the book structure and function are closely correlated. The two e…
Extending the hosts of Tectiviridae into four additional genera of Gram-positive bacteria and more diverse Bacillus species
2018
Tectiviridae are composed of tailless bacteriophages with an icosahedral capsid and an inner membrane enclosing a double-stranded 15 kb linear DNA genome. Five of the seven previously studied Tectivirus isolates infect bacteria from Bacillus cereus sensu lato group (Betatectivirus), one distantly related member (PRD1) infect Enterobactericeae (Alpatectivirus) and one recently discovered virus infect Gluconobacter cerinus (Gammatectivirus). Here we expand the host spectrum of Betatectivirus elements to four additional genera (Streptococcus, Exiguobacterium, Clostridium and Brevibacillus) and to more distantly related Bacillus species (B. pumilus and B. flexus) by studying the genomes of four…
Success factors of game industry crowdfunding campaigns
2017
Tätä mallia testattiin haastattelemalla kolmen eri joukkorahoituskampanjan perustajia. Aiemman teorian sekä haastattelujen perusteella selvisi, että pelialan joukkorahoituskampanjoilla on kuusi selkeää menestystekijää: kampanjan palkkiot, kampanjan tavoitteet, kampanjan sekä tuotteen laatu, projektitiimi, joukkorahoittajien luoma yhteisö sekä suoritetut esivalmistelut. Tutkimustuloksia ja niiden perus…
